白血病患者WT1 基因表达及其临床意义

摘要: 本文探讨WT1 基因在各种白血病中的表达及其临床意义。应用逆转录酶链反应(RT2PCR) 法测定K562 细胞株,23例白血病患者,其中AML12 例,ALL3 例,CML7 例,CLL1 例;2 例恶性淋巴瘤( Ⅲ期) 患者骨髓或外周血及10 例正常人外周血WT1 基因表达。K562 细胞株高度表达WT1 ,急性髓性白血病(AML) 12 例中10 例,急性淋巴细胞白血病(ALL) 3 例中2 例,急性粒细胞白血病(CML) 急性变6 例表达阳性;CML 加速期、慢性淋巴细胞白血病、恶性淋巴瘤( Ⅲ期) 和10 例正常人外周血T1 基因表达阴性。WT1 基因在早期造血细胞中表达,与急性白血病密切相关,有可能作为残余白血病细胞检测的特异性指标。

Abstract: To elucidate the expression of Wilms’tumor gene (WT1) in various types of leukemia and its clinical implication. WT1 mRNA was detected in K562 cell line , 23 leukemia patient s (12AML patient , 3 ALL patient , 7 CML patient , 1 CLL patient . ) 2 lymphoma patient s in three stage , and 10 healthy subjects by using RT2PCR. WT1 mRNA expression was revealed in K562 cell line , 10 of 12 AML patient s , 2 of 3 ALL patients , 6 CML patient s in blast crisis. WT1 mRNA was undetecatable in 1 CML patient in accelerated , 1 CLL patient , 2 NHL in three phase and 10 healthy subject s. WT1 gene is expressed in early hematopoietic cells , it is closely related with acute leukemia , and supposed to be a new marker for monitoring MRD in acute leukemia.
