彗星试验检测间接诱变剂对小鼠睾丸细胞的DNA 损伤

STUDY ON DNA DAMAGE INDUCED BY INDIRECT MUTAGENS USING COMET ASSAY IN TESTICULAR CELLS IN VITRO

摘要: 目的:探讨组织原代细胞在检测间接诱变剂中的作用。方法:采用单细胞凝胶电泳试验(彗星试验) 对3 种典型的间接诱 变剂(环磷酰胺、2 - 氨基芴和二甲基苯蒽) 所致的离体小鼠睾丸细胞的DNA 损伤进行了研究。结果:在一定浓度范围内,3 种 受试物均可直接引起离体小鼠睾丸细胞的DNA 单链断裂,出现拖尾的彗星细胞。其拖尾细胞百分率和DNA 迁移长度均随 受试物浓度的增加而增加,且有明显的剂量反应关系。结论:原代小鼠睾丸细胞具有一定的代谢活化作用,可以用于彗星试验 直接检测间接诱变剂所致的DNA 损伤研究。

Abstract: Purpose :To stuty the metabolic role of primary tissue cells in detecting indirect mutagen. Methods the DNA damage effect s of cyclophosphamide , dimethylbenz (a) anthracene and 2 - aminofluorene were studied using the single cell gel elect rophoresis technique (Comet assay) in isolated mice testicular cells. Results : In certain concent ration , the three tested substances could induce the breakage of DNA single st rand and resulted in comet cells with tail. Both the number of comet cells and the length of DNA migration increased with increasing of concent ration of tested substances. Conclusion : The primary mice testicular cells had certain metabolic role and could be used to detect the effect s of DNA damage induced by indirect mutagen in comet assay.
