hTR基因反义核酸对K562 细胞端粒酶活性和凋亡的影响

摘要: 目的: 研究人类端粒酶(hTR) 基因的反义核酸对K562 细胞端粒酶活性和细胞凋亡的影响。方法:采用TRAP 法检K562 细胞在反义核酸处理前后端粒酶活性的变化,以及采用TUNEL 法观察反义核酸处理前后细胞凋亡的变化。结果: hTR基因反义核酸能有效抑制K562 细胞的端粒酶活性,并且诱导K562 细胞发生凋亡。结论:hTR 基因反义核酸能显著抑制肿细胞的端粒酶活性,为恶性肿瘤治疗提供了一条新的有效的途径。

Abstract: Purpose : To study the inhibitory effect of human telomerase activity and the apoptosis by an antisense oligonucleotide against human telomerase RNA ( hTR) in K562 cells. Methods : Telomerase activity of K562 cells with antisense t reatment was measured using the TRAP assay , and apoptosis was studied by means of TUNEL method. Results : The telomerase activity was effectively inhibited , and the apoptosis was induced by the antisense t reatment . Conclusion : Antisense oligonucleotide against hTR can effectively inhibit the telomerase activity of K562 cells.
