摘要: 背景与目的: 利用报告基因监测肝干细胞向胰腺细胞的转分化潜能。 材料与方法: 通过PCR从小鼠基因组中克隆胰十二指肠同源盒基因-1(Pancreatic duodenal homeobox-1, PDX-1)启动子片段,构建PDX-1启动子调控的绿色荧光蛋白报告载体(pPDX-1 EGFP),并利用报告载体标记的肝原始细胞系(Liver epithelial progenitor cells, LEPCs)观察肝干细胞向胰腺细胞的转分化潜能。 结果: 获得PDX-1启动子调控的绿色荧光蛋白报告载体及报告载体标记的LEPCs。 结论: pPDX-1 EGFP报告载体的成功构建为研究LEPCs向胰腺细胞的转分化提供了简捷的分子工具。

Abstract: BACKGROUND & AIM: To construct the fluorescence protein expression vector as a reporter for studying the transdifferentiation potency of liver epithelial progenitor cells (LEPCs) to pancreas-like cells. MATERIAL AND METHODS: The Pancreatic duodenal homeobox-1(PDX-1) promoter was amplified by PCR from mouse tail genomic DNA and subcloned into promoter-removed pEGFP N1 vector. By using the reporter gene tagging cells, the phenotype variation of LEPCs transdifferentiated into pancreas-like cells could be evaluated. RESULTS: EGFP expression vector driven by PDX-1 promoter could monitor the transdifferentiation of LEPCs into pancreas-like cells in a real-time manner. CONCLUSION: The reporter vector is useful for studying the transdifferentiation of LEPCs into pancreas-like cells.
