对苯二酚诱发果蝇生殖细胞非整倍体的研究

ANEUPLOID INDUCTION BY HYDROQUINONE IN GERM CELLS OF DROSOPHILA MELANOGASTER

摘要: 本研究以黑腹果蝇Oregan K品系评价了有丝分裂抑制剂对苯二酚(HQ) 在果蝇生殖细胞中的非整倍体诱发效应,秋山仙素(COL) 为本试验的阳性对照。羽化后1 —2 天的雌雄成虫口饲染毒后,分别与未染毒个体交配,分析F1 代中例外子代的频率。结果发现:HQ 在三个高剂量组(12 - 18mg/ ml) 均显著诱发果蝇生殖细胞非整倍体,使XXY及XO 例外子代频率显著升高( P < 0105) ,COL 于所有受试组(215 —20μg/ ml)中,均显著诱发生殖细胞非整倍体( P < 0. 01 —0105) 。试验表明:HQ 及COL 均能够在果蝇生殖细胞形成过程中,导致性染色体的丢失和不分离,但HQ 作用效应远低于COL ;雌性果蝇对受试物的敏感度明显低于雄性果蝇。

Abstract: The Oregon K system of Drosophila melanogaster was employed for the detection of germ cell aneuploid induced by hydroquinone (HQ) and colchicine (COL) . Adult flies were treated by feeding the chemicals. HQ at doses 12218mg/ ml and COL (2. 5 - 20μg/ ml) significantly induced the exceptional off spring (XXY, XO) derived f rom aneuploidy germ cells. It was showed than COL had much higher efficiency on aneuploid induction than HQ and shared a strong effect on chromosome lossing in treated parental males. No significant sex differences among HQ t reated animals for the chromosome lossing and mal2segregation in germ cell development were found.The experiment indicated that HQ and COL are aneugens in Drosophila germ cells.
