莲房原花青素对黑色素瘤B16细胞的诱导分化作用

摘要: 背景与目的: 探讨莲房原花青素(LSPC)对荷瘤小鼠体内黑色素瘤B16细胞的诱导分化作用。 材料与方法: 以荷黑色素瘤B16小鼠为模型,随机分LSPC 3个剂量(60、120、240 mg/kg)组和阴性对照组,每组10只,实验组用LSPC、阴性对照组用等体积溶剂分别灌胃13 d后取材,检测黑色素瘤B16细胞中酪氨酸酶活力、黑色素含量、S_100蛋白表达,并用光学显微镜和透射电镜观察其细胞形态的变化。 结果: 60~120 mg/kg的LSPC能抑制黑色素瘤B16细胞在小鼠体内生长,且呈浓度依赖关系。其中,120 mg/kg LSPC组作用最显著(P<0.01),抑瘤率达55.3%,并使B16细胞内酪氨酸酶活力和黑色素含量分别提高28.5%和23.8%,与阴性对照比较,差异有统计学意义(P<0.05),并使S_100蛋白表达下降。 结论: LSPC显著抑制B16细胞在小鼠体内的生长,并从形态上和功能上诱导B16细胞分化。

Abstract: BACKGROUND & AIM: To investigate the differentiation of mice B16 melanoma induced by proanthocyanidins from lotus seedpod (LSPC)in vivo. MATERIALS AND METHODS: B16 melanoma_bearing mice were used to study the inhibitory effect of LSPC on B16 melanoma, the effect of LSPC on tyrosinase activity, and the ability of melanin formation, and the morphology change of B16 melonoma and the expression of S_100 protein. RESULTS: LSPC(60-120 mg/kg)could dose_dependently inhibit the growth of B16 melanoma,The effects in the group with dosage of 120 mg/kg was the most marked. The inhibition rate of B16 melonoma was 55.3%, while the activity of tyrosinase, the content of melanin increased 28.5% and 23.8%,respectively. There were significant differences compared with control group(P<0.05). The expression of S_100 protein was down_regulated. CONCLUSION: LSPC could significantly inhibit the growth of mice B16 melanoma cells, and induce the differentiation of B16 melanoma cell in morphology and function.
