茶树槲寄生“螃蟹脚”水提物的急性毒性和遗传毒性

Acute toxicity and genotoxicity of crude water extracts from tea mistletoe Viscum Liquidambaricolum Hayata

摘要: 目的:研究“螃蟹脚”水提物的经口急性毒性和遗传毒性。方法:采用大鼠经口急性毒性试验、Ames试验、小鼠骨髓嗜多染红细胞微核试验和小鼠精原细胞染色体畸变试验对“螃蟹脚”水提物的安全性进行评价。结果:经口急性毒性试验表明,大鼠灌胃给予“螃蟹脚”水提物,其半数致死量约为21.5 g/kg,因此“螃蟹脚”水提物属于实际无毒物。Ames试验、小鼠骨髓嗜多染红细胞微核试验和小鼠精原细胞染色体畸变试验结果均为阴性。结论:在本次实验条件下,“螃蟹脚”水提物属于实际无毒级别,而且未观察到遗传毒性。

Abstract: OBJECTIVE: To investigate the acute toxicity and genotoxicity of crude water extracts from tea mistletoe (CWE). METHODS: The following assays were used for the investigation:Ames assay, acute toxicity in rats, micronuclei of mouse bone marrow polychromatic erythrocytes, and mouse spermatogonium chromosome aberrations. RESULTS: Oral acute toxicity test revealed that the LD50 of CWE was approximate 21.5 g/kg in rats, thus CWE was relatively non-toxic. No mutagenic activities were detected in the Salmonella typhimurium reverse mutation assay (Ames test), both with and without metabolic activation. In addition, CWE demonstrated no positive adverse effects in the in vivo micronucleus test or spermatogonium chromosome aberration test of ICR mice. CONCLUSION: Under our experimental conditions, water extracts from CWE showed no evidence of acute toxicity and no positive evidence of genotoxicity.
