贝伐单抗动脉介入超选化疗对大鼠脑胶质瘤的影响

Experimental transcarotid artery chemotherapy on gliomas

摘要: 目的:探讨贝伐单抗动脉介入超选化疗对大鼠脑胶质瘤的影响。方法:移植脑胶质瘤大鼠模型建立成功后,随机分成对照组(经尾静脉注射等量生理盐水)、静脉注射顺铂组(V+CDDP)、静脉注射贝伐单抗组(V+BVZ)、动脉注射顺铂组(A+CDDP)、动脉注射贝伐单抗组(A+BVZ),给药(均为5 mg/kg)7 d后测量胶质瘤体积,并采用Real time-PCR和Western blot检测脑胶质瘤组织血管内皮生长因子(VEGF)、P21、Bcl-2和金属基质蛋白2(MMP-2)的mRNA和蛋白表达情况。结果:A+CDDP组肿瘤体积[(5.58±0.37)mm3]较V+CDDP组小[(7.67±0.70)mm3],A+BVZ组肿瘤体积[(4.81±0.20)mm3]较V+BVZ组小[(6.57±0.69)mm3],差异均具有统计学意义(P均 < 0.01)。同时,V+BVZ组肿瘤体积显著小于V+CDDP组(P < 0.01),A+BVZ组肿瘤体积显著小于A+CDDP组(P < 0.05),与静脉注射相比,动脉注射组VEGF、Bcl-2和MMP-2 mRNA和蛋白的表达均显著降低(P < 0.05),P21 mRNA和蛋白的表达显著升高(P < 0.05)。与注射CDDP相比,注射BVZ组VEGF、Bcl-2和MMP-2 mRNA和蛋白含量均显著降低(P < 0.05),P21 mRNA和蛋白表达均显著升高(P < 0.05)。结论:应用BVZ动脉介入超选化疗,能有效控制脑胶质瘤的大小,从而达到较好的治疗效果。

Abstract: OBJECTIVE: To investigate the effectiveness of transcarotid artery chemotherapy on gliomas in rats. METHODS: After the brain glioma model was established successfully,rats were randomly divided into control group(the same amount of saline injected through the tail vein),intravenous injection of cisplatin group (V+CDDP),intravenous bevacizumab group (V+BVZ),arterial injection of cisplatin group (A+CDDP),arterial injection of bevacizumab group (A+BVZ). After treatment for 7 d,the glioma volume was detected,and the expression of VEGF,P21,Bcl-2 and MMP-2 protein and mRNA were detected by Western blot and real time-PCR. RESULTS: The glioma volume in A+CDDP group[(5.58±0.37) mm3] was smaller than that in V+CDDP group[(7.67±0.70) mm3]. The glioma volume in A+BVZ group[(4.81±0.20) mm3] was smaller than that in V+BVZ group[(6.57±0.69) mm3]. Differences were statistically significant (P < 0.01),and it was shown that artery injection had a strong inhibitory effect on the size of glioma. At the same time,the glioma volume in V+BVZ group was smaller than that in V+CDDP group (P < 0.01),the glioma volume in A+BVZ group was smaller than that in A+CDDP group (P < 0.05). BVZ had a stronger inhibitory effect on glioma size compared with CDDP,and artery injection had a stronger inhibitory effect than intravenous injection on the size of glioma. Compared with intravenous injection,the expression of VEGF,Bcl-2 and MMP-2 protein and mRNA were significantly decreased (P < 0.05) and the expression of P21 protein and mRNA was significantly increased in arterial injection group (P < 0.05). Compared with CDDP,the expression of VEGF,Bcl-2 and MMP-2 protein and mRNA were significantly decreased (P < 0.05) and the expression of P21 protein and mRNA was significantly increased (P < 0.05) in BVZ group. CONCLUSION: Application of BVZ transcatheter arterial chemoinfusion effectively inhibited the size of glioma,therefore,this approach may be useful for providing better therapeutic effect.
