电离辐射对人乳腺癌MCF7 细胞系hR24L 基因表达的影响

摘要: 目的:研究电离辐射对人类DNA 修复基因hR24L 表达的影响。方法:通过RT - PCR 从人乳腺癌MCF7 细胞总RNA 中扩增hR24L 的开放阅读框(ORF) ,经测序无突变后用γ射线照射细胞。照射后不同时间收集细胞,提取总RNA ,用Northern印迹杂交分析hR24L 表达变化。结果:hR24L 基因的转录在电离辐射后1 h 后开始上升,2 h 达高峰,3 h 恢复正常。结论:该基因的表达具有损伤诱导性特点,对γ射线损伤有修复作用。

Abstract: Purpose : To study the effect of radiation on the expression of human DNA repair gene hR24L. Methods : We amplified it s open reading f rame f rom human breast cancer MCF7 cells by RT2PCR , and had its sequence checked , irradiated the cells with gamma2ray at a total dose of 5 Gy followed by incubating the cells for different time , collecting cells , isolating the total RNA , analysed hR24L2mRNA by Northern bloting. results :The expression of hR24L2mRNA began to increase at about one hour after irradiation and reached the maximum at about two hours after irradiation. Conclusion : The expression of hR24L gene is damage2inducible , this expression can repair the damage caused by gamma2ray irradiation.
