载脂蛋白E基因多态性和帕金森病的相关性

Association between Polymorphism of Apolipoprotein E Gene and Parkinson's Disease

摘要: 背景与目的: 探讨乌鲁木齐地区人群中载脂蛋白E(ApoE)基因多态性与帕金森病(Parkinson's disease, PD)的相关性。材料与方法: 用PCR- RFLP方法检测51例散发性PD和52例正常对照的ApoE基因型。根据有无痴呆将所有PD患者分为二组,即PDD组和PD组。用χ2检验进行关联分析。 结果: PD+PDD组、PD组和PDD组与对照组间的ApoE基因型频率差异无显著性(P > 0.05)。PD组ApoE等位基因频率与对照组相近。PD+PDD组和PDD组的ε 2和ε 3频率类似于对照组,但是,ε 4频率(分别为12.8 %和16.7 %)明显高于正常对照组(3.9 %),组间比较差异有显著性(P < 0.05)。结论:ApoE ε 4等位基因对PD的发生无明显影响,但是,它是伴有PD的痴呆患者的发病危险因素。

Abstract: BACKGROUND & AIM: To explore the relationship between polymorphism of apolipoprotein E(ApoE) gene and the risk of Parkinson's disease(PD) in the population of Urumqi region. MATERIAL AND METHODS: PCR and RFLP were used to detect ApoE genotypes in 51 sporadic PD cases and 52 healthy elderly persons as controls. The whole PD patients were categorized as the PDD group and the PD group according to presence or absence of dementia . Association analysis was performed using χ2 test. RESULTS: Genotype frequencies of ApoE in PD+PDD mixed group, PD group and PDD group did not differ significantly from that in controls(P>0.05). Allele frequency of ApoE in PD group was similar to that in controls. There were no differences in the frequencies of ε 2 and ε 3 alleles in PD+PDD, PDD groups and controls. However, the frequencies of ε 4 allele in PD+PDD and PDD groups(12.8 % and 16.7 %, respectively) were obviously higher than that in controls, and the differences were statistically significant(P<0.05). CONCLUSION: ApoE ε 4 allele does not have a marked effect on the development of PD, but may be a genetic risk factor for dementia with PD in the patients studied.
