STUDY ON SCID MOUSE MODEL OF HUMAN GASTRIC CANCER CONSTRUCTED USING ORTHOTOPIC IMPLANTATION AND THEIR BIOLOGICAL PROPERTIES

摘要: 将传代于裸鼠皮下的人胃癌SGC - 7901 完整组织原位移植于SCID 鼠,探讨移植后自主生长的肿瘤生物学特性及其侵袭转移与形态学特征。移植成功率为100 %。移植瘤保持了人胃癌选择素表达的生物学特性并显示人胃癌侵袭转移的体内表现。表明移植部位的微环境对移植瘤的生物学行为有一定影响。原位移植后自主生长的肿瘤较皮下移植模型更客观地模拟了患者体内的侵袭转移过程,可适用于人类胃癌的实验治疗研究和为进一步探讨粘附分子介导的胃癌侵袭转移机制提供有效工具。

Abstract: Histologically intact tissue of human gast ric carcinoma SGC - 7901 maintained by passage in the subcuitis of nude mouse was implanted orthotopically into the stomach wall of SCID mouse. Their biological properties ,ways of invasion and metastasis and morphological characteristic were studied , All mice with t ransplanted tumours exhibited 100 % t ransplantability and metastatic ability ,as well as various manifestation reminescent of tumour behaviour in patients. These included :local growth ,regional invasion ,spontaneous metastasis to lymph nodes and liver ,and peritoneal seeding. RT2PCR confirmed that t ransplanted tumours had identical characteristic of expression of cell adhesion molecules P2selectin as f resh human specimens of gast ric cancer. This indicated that the microenvironment in the t ransplantation site had certain influience on the biological behaviour of the t ransplahted tumours which grew independentely in the stomach of SCID ,making a better model than that of subcutaneously t ransplanted tumours for futther study on invasion and metastasis mediated by adhesion molecules and experimental therapy further of human gast ric cancer.
