镉对NRK肾细胞PKCα表达的影响及PKC抑制剂BisⅠ的生物学作用

Effects of cadmium chloride on the expression of PKCα in NRK cells and the biological effect of PKC inhibitor

摘要: 目的: 探讨不同浓度氯化镉(CdCl2)对NRK细胞内蛋白激酶Cα(PKCα)mRNA及其蛋白质表达的影响,以及PKC抑制剂Bisindolymaleimide I (Bis I)对镉染毒NRK细胞PKCα mRNA及蛋白质表达的生物学作用。 方法: 应用实时荧光定量PCR技术检测不同浓度( 0、1.25、2.50、5.00、10.00 μmol/L) CdCl2对NRK肾细胞处理6 h与12 h后PKCα mRNA表达的影响,同时用Western blot技术检测CdCl2对NRK肾细胞内PKCα蛋白质表达的影响。用PKC抑制剂Bis I预处理细胞后,用不同浓度CdCl2(0、2.50、5.00 μmol/L)进行染毒,再检测PKCα mRNA及蛋白质的表达变化。 结果: CdCl2浓度在1.25~10.00 μmol/L范围内,可引起NRK肾细胞内PKCα mRNA及蛋白质表达水平升高,并呈时间-剂量-效应关系;PKC抑制剂Bis I能明显抑制镉引起的PKCα mRNA及蛋白质的表达。 结论: CdCl2可以诱导NRK肾细胞内PKCαmRNA及蛋白质表达,PKC抑制剂Bis I具有一定程度抑制CdCl2对PKCα的诱作用,因此在抑制CdCl2对生物机体的伤害作用方面具有生物学意义。

Abstract: OBJECTIVE: To evaluate the effect of cadmium chloride (CdCl2)on the expression of PKCα mRNA and protein in NRK cells as well as the biological effect of PKC inhibitor(Bisindolymaleimide I) on cells treated with cadmium. METHODS: Real-time fluorescence quantitative PCR assay was applied to test the effects of CdCl2 on the expression of PKCα mRNA in NRK cells exposed to CdCl2 at various concentrations (0, 1.25, 2.5, 5 and 10 μmol/L). Western blot was used to measure the effects of CdCl2 on the expression of PKCα protein in NRK cells exposed to CdCl2 at the same concentrations. Additionally, the expressions of PKCα mRNA and protein were also assessed in NRK cells exposed to CdCl2 after treated by PKC inhibitor. RESULTS:The expressions of PKCα mRNA and protein in NRK cells exposed to CdCl2 at the concentrations between 1.25 and 10 μmol/L were increased, and a dose-effect relationship was found. PKC inhibitor had an obvious effect on the expressions of PKCα mRNA and protein in the cells after CdCl2 treatment. CONCLUSION:CdCl2 could induce an increase in expressions of PKCα mRNA and protein, and the PKC inhibitor showed an obvious inhibitory effect.
