流式细胞术测定COX-2,iNOS基因蛋白在食管癌上的表达及其与Bcl-2,PCNA的相关性

摘要: 背景与目的: 研究环氧化酶-2(Cyclooxygenase 2,COX-2),Ⅱ型一氧化氮合酶(TypeⅡnitric oxide synthase,NOS2)基因蛋白在食管鳞癌上的表达及与食管癌的分化和淋巴结转移的关系,并探讨其与PCNA、Bcl-2的相关性。材料与方法: 采用流式细胞术定量检测65例食管鳞状细胞癌上增殖细胞核抗原、iNOS、PCNA及Bcl-2基因的蛋白表达量(用荧光指数FI表示)。 结果: COX-2,iNOS在低分化型(Ⅲ级)鳞癌上的表达量明显高于分化型(Ⅰ、Ⅱ级)鳞癌(其P值分别为0.000 1,0.038 5);二者之间差异有显著性。该两种基因蛋白的表达强弱均与淋巴结转移无关。COX-2与PCNA、Bcl-2呈明显正相关,而iNOS与PCNA、Bcl-2无明显相关性。 结论: COX-2,iNOS的表达强弱与食管癌的分化密切相关 ,二者在促食管癌的发生发展上可能有互相协同作用;COX-2可能促进食管癌细胞的增殖,并通过刺激Bcl-2蛋白表达抑制凋亡。

Abstract: BACKGROUND & AIM: To study the expressions of Cyclooxygenase 2(COX-2),TypeⅡ nitric oxide synthase(iNOS) proteins in esophageal squamous cell cancer(ESCC) and their relationship to tumor differentiation,lymph nodes metastasis,Proliferating cell nuclear antien(PCNA) and Bcl-2. MATERIAL & METHODS: The expressions of COX-2,iNOS, PCNA and Bcl-2 proteins in 65 cases of ESCC were defermined quantitatively by Flow Cytometry. RESULTS: The expressions of COX-2,iNOS in poorly-differentiated ESCC were higher than those of well- differentiated ESCC(P=0.0001,0.0385).There was positive correlationship between COX-2 and iNOS. The expressions of the two proteins did not correlate with lymph nodes metastasis.COX-2 expression correlated with PCNA,Bcl-2 expressions .No significant association was found between iNOS expression and PCNA,Bcl-2 expressions. CONCLUSIONS: The expressions of COX-2 and iNOS closely related to tumor differentiation in ESCC.They may promote carcinogenesis in coordination.COX-2 may contribute to tumor proliferation,suppress apoptosis by stimulating Bcl-2 expression.
