酒精对大鼠睾丸的亚慢性氧化损伤及维生素A 的保护作用

Oxidation Injury in Rats Testis Induced by Chronic Alcohol Intake and the Protective Effect of Vitamin A

摘要: 背景与目的: 观察亚慢性摄入酒精对大鼠睾丸结构和功能的损害及维生素A(Vitamin A,VA)的保护作用。材料与方法: 30只健康雄性成年SD大鼠随机分为对照组、酒精组、酒精+VA组,各组每日分别灌胃给予酒精0、7.5 g/kg、酒精7.5 g/kg+VA 50 μg/kg,连续13周后对各组大鼠的精子计数、精子活动率、精子畸形率、血清睾酮(Serum testosterone,T)进行检测,光、电镜观察睾丸生精上皮的组织学改变。同时测定睾丸线粒体中丙二醛(Malonaldehyde MDA)的产生量。结果: 与对照组相比,酒精组大鼠精子计数减少,精子活动率下降,精子畸形率升高,血清T水平明显降低,睾丸生精上皮结构破坏,支持细胞和各级生精细胞均有退化变性,睾丸线粒体丙二醛含量明显升高;酒精+VA组较单纯酒精组精子计数、精子活动率有所上升,生精细胞退化变性程度减轻,睾丸线粒体MDA减少,但血清T仍低于对照组。结论: 亚慢性摄入酒精抑制精子生成和睾酮合成,补充VA可以限制酒精引起的睾丸过氧化损伤,保护睾丸的生精功能,但是仍有间质细胞合成睾酮障碍。

Abstract: BACKGROUND & AIM: To study the effects of chronic alcohol consumption on testicular structural and functional disturbance in male rats and the protective effect of vitamin A against it. MATERIAL AND METHODS: Thirty 30 healthy Sprague-Dawley adult male rats were randomly divided into 3 groups: control, alcohol 7.5 g/kg and alcohol 7.5 g/kg+vitamin A 50 μg/kg. Alcohol and vitamin A were administrated to the rats for 13 weeks by a gastric tube. The sperm counting,motility and the percentage of abnormal sperm were observed. Serum testosterone (T) was determined. The patholgical changes of testicle tissue of rats were observed by light and electron microscopy. The malonaldehyde (MDA) content of testicular mitochondrial was simultaneously determined. RESULTS: Compared with control group the sperm counting and motility of the alcohol group decreased (P<0.05). The frequency of abnormal sperm increased (P<0.05). Serum T level significantly decreased (P<0.05). The MDA content was markedly higher than that of the control (P<0.05). Histological evaluation of testes revealed that seminiferous epithelium was disorganized and that sertoli cells and germ cells degenerated in alcohol-treated rats. Vitamin A supplementation inhibited the testicular mitochondrial MDA formation and as a result germ cells degeneration was improved. Sperm counting and motility were higher than that of the alcohol treated group,but it didn't prevent the decline of serum T level. CONCLUSION: Chronic alcohol intake led to spermatogenesis and steroidogenesis inhibition in testes. Vitamin A supplementation prevented testis from the peroxidation injury which was a result of chronic alcohol exposure. To a certain degree, vitamin A supplementation protected the spermatogenesis function, but Leydig cell injury still occurred.
