转人TNFα基因对肾细胞癌细胞致瘤性的影响

INFLUENCE ON THE TUMORIGENICITY OF THE HUMAN RENAL CELL CARCINOMA ( RCC) CELLS TRANSDUCED WITH HUMAN TNFα GENE

摘要: 目的:观察转人TNFα基因对肾细胞癌细胞(RCC) 致瘤性的影响,为进一步对RCC 的基因治疗打下基础。方法:将人TNFα基因构建入逆转录病毒载体,经包装、鉴定后,感染RCC 细胞株78620 , EL ISA 法测转人TNFα基因78620 细胞上清中TNFα的活性,同时,将转人TNFα基因的78620 细胞接种裸鼠,观测转TNFα前后RCC 细胞致瘤性的变化。结果: 转人TNFα78620 细胞上清TNFα的浓度平均为(5 004 ±624) pg/ ml ,接种裸鼠后无肿瘤长出。结论:转人TNFα基因使建株RCC 失去了致瘤性。

Abstract: Purpose : To study the influence on the tumorigenicity of the human renal cell carcinoma (RCC) cells by t ransfection of human TNFαgene. Methods : hTNFαcDNA was inserted into ret roviral vector to const ruct recombinant vector pL ( TNFα) SN. The packaged and confirmed ret rovirus were infected to RCC cell line.Then , nude mice were injected by 78620 cell infected with the hTNFαgene or not , and the tumorigenecity of RCC was observed. Results : The mean expression level of TNFαin the supernatant of TNFα2t ransfected 78620 cell2culture medium was (5 004 ±624) pg/ ml. And the nude mice injected with TNFα gene engineered RCC cells had no tumor growth. Conclusion : The TNFαgene engineered human RCC cells lost tumorigenicity.
