Consumption of transgenic rice with the cry1Ab/Ac gene on intestinal mucosa, intraepithelial lymphocytes and goblet cells in Cynomolgus monkey

摘要: 目的:研究转cry1Ab/Ac基因大米对食蟹猴小肠黏膜形态及上皮内淋巴细胞和杯状细胞数量的影响。方法:以转cry1Ab/Ac基因大米为受试物,选用24只食蟹猴,雌雄各半,随机分成3组,即普通大米对照组、转cry1Ab/Ac基因大米低、高剂量组。对照组、低剂量和高剂量组给予的猴饲料中转cry1Ab/Ac基因大米含量分别为0、17.5%和70%。每天饲喂2次,每次(100±10)g,持续6个月,恢复期4周。分别进行一般毒性指标的检测:临床症状观察、体质量、体温、心电图、血液学、血清生化、尿生化、脏器质量以及组织病理学检查。解剖取十二指肠、空肠、回肠进行HE染色和PAS染色,观察小肠黏膜形态、上皮内淋巴细胞及杯状细胞数目。结果:饲喂6个月后,与普通大米对照组相比,转cry1Ab/Ac基因大米低剂量组和高剂量组食蟹猴的一般常规检查未见异常。小肠黏膜形态各组均未见病理改变,低、高剂量组的上皮内淋巴细胞和杯状细胞数与对照组相比差异均无统计学意义(P>0.05)。结论:饲喂转cry1Ab/Ac基因大米6个月对食蟹猴小肠黏膜形态、上皮内淋巴细胞及杯状细胞数量未见不良影响。

Abstract: OBJECTIVE: To investigate the effect of transgenic rice with the cry1Ab/Ac gene on morphology of intestinal mucosa,cell number of intraepithelial lymphocytes and goblet cells in Cynomolgus monkey. METHODS: Twenty-four cynomolgus monkeys,half male and female,were randomly divided into 3 groups including control group of ordinary rice,low dose group and high dose groups of transgenic rice with the cry1Ab/Ac gene. Monkeys were fed twice a day and each feeding was (100±10) g for 6 months with 4 weeks recovery period. Monkeys were observed for general toxicity,clinical effects,body weight,body temperature,electrocardiogram,hematology,serum biochemistry, urinary biochemistry,organ weight and histopathological examination. HE and PAS staining were done for duodenum, jejunum and lleum. The morphology of intestinal mucosa,cell number of intraepithelial lymphocytes and goblet cells were observed under microscope. RESULTS: After six months,detection of transgenic rice with the cry1Ab/Ac gene were 0, 17.5% and 70%,in control,low dose and high dose groups,respectively. Compared with the control group,no abnormalities in toxicity,etc. were found in the treated groups. No pathological changes were observed in intestinal morphology. Cell number of intraepithelial lymphocytes and goblet cells were not statistically different between control and treated groups. CONCLUSION: Transgenic rice with the cry1Ab/Ac gene had no side effects on small intestine mucosal morphology,cell number of intraepithelial lymphocytes and goblet cells in Cynomolgus monkey.
