格兰西隆的致突变性研究

STUDIED ON MUTAGENICITY OF GRANISETRON

摘要: 应用Ames 试验,体外染色体畸变试验和微核试验对格兰西隆进行了致突变性研究。研究结果,格兰西隆的各剂量组对TA98 ,TA97 ,TA100 ,TA102 在加和不加S9mix 条件下均无致突变性,染色体畸变试验中,在加S9mix 条件下,15μg/ ml 和30μg/ ml 剂量对CHL 细胞诱发的畸变率分别为6 %和7 % ,为阳性。以418mg/ kg 剂量以上对小鼠骨髓多染红细胞有诱发微核作用。实验结果提示,格兰西隆具有潜在致突变性。

Abstract: The Mutageicity of Graniset ron was studied by Ames test , chromosome aberration assay of mammlian cell (CHL) in vit ro and mouse marrow micronucleus assay. The results showed that Graniset ron had no mutagenicity on salmonella typhimurium st rain TA97 , TA98 , TA100 and TA102 with or mithout S9 mix. Chromosome aberration rates induced by 15μg/ ml and 30μg/ ml doses with S9 mix were 6 % and 7 % respectively , Possible postive. The freqaency of male mice micronuleus rate induced by 4. 8mg/ kg dose were signifioant disfereonces as Compared with negative cont rocl ( P < 0. 01) , The result s showed that Graniset ron more than 4. 8mg/ kg Graniset ron Could also induce mouse marrow micronucleus rates increase , So. Graniset ron is mutagenic.
