EFFECTS OF VANADIUM PENTOXIDE ON RAT EMBRYO MIDBRAIN CELL PROLIFERATION AND DIFFERENTIATION

摘要: 本研究采用微团培养观察了V2O5 对大鼠胚胎中脑细胞增殖和分化的影响,旨在为进一步揭示其发育毒作用机理提供依据。结果显示,V2O5 对体外中脑细胞的增殖和分化均有抑制作用,并有明显的剂量- 反应关系。V2O5 抑制中脑增殖和分化的I C50 分别为911 和618μmol/ l 。体内/ 体外结合试验显示,V2O5 在210mg/ kg 时对细胞分化有明显的抑制作用,而在510mg/ kg 时才对细胞增殖出现抑制作用,按照Renault 的分类标准,可判定V2O5 是胚胎中脑细胞的特异抑制物。

Abstract: Vanadium is a proven animal teratogen which can induce dilation of latermal vent ricles and skeletal abnormal in rat , although the mechanism of it s developmental toxicity is unknown. In this paper , the micromass rat embryo midbrain cell culture was used in a affort to determine potential mechanisms by which Vanadium Pentoxide (V2O5) may exert it s teratogenic effect . In vit ro test , a dose2dependent inhibition of differentiation and proliferation of midbrain cells were observed. The 50 % - inhibitory concent ration ( IC50) for cell differntiation and cell survial or proliferation 50 % the cont rol value ( IC50) are 6. 8 and 9. 1μmol/ L , respcectively. In vivo/ in vit ro test , V2O5 could inhibit cell differentiation at 2. 0 mg/ kg body weight , but inhibit cell surival or proliferation at 5. 0mg/ kg. According to class standard of Renault , el al (1989) , V2O5 may be a specific inhibitor of embryonic midbrain cell.
