燃煤烟颗粒介导的DNA 氧化损伤 I. 对脱氧核糖羟化降解的影响

THE OXIDATIVE STRESS MEDIATED BY THE SMOKE TAR FROM BURNING COAL-THE HYDROXYLATIVE DAMAGE TO DEOXYRIBOSE

摘要: 煤厂的煤烟颗粒有机溶剂(硝基甲烷和二氯甲烷) 和水相二种类型的抽提物都能有效增强、加速Udenfriend 反应体系中脱氧核糖(dR) 羟化降解,并呈现一定的剂量一效应关系。用脱氧腺苷(dA) 替代,也能获此结果。而燃油厂的油烟颗粒则无上述作用。经ESR 测定,燃煤厂的煤烟颗粒自由基g 值为210016 ;燃油厂的油烟颗粒自由基g 值为211008 ,前者接近于半醌自由基,后者为类似于大气飘尘中性质完全不同的宽谱、稳定自由基。提示煤烟的诱变性和致癌性与其对OH 生成的加速作用及其对DNA 造成的氧化损伤有一定关系。

Abstract: Both the organic and aqueous ext ract s of smoke tar f rom burning coal could accelerate hydroxylative degradation of deoxyribose (dR) of deoxyadenosine (dA) in Uderf riend reaction system , and showed dose2effect relationship to some extent . However , smoke tar f rom burning oil showed none of these effect s. Free radicals were determined by ESR(elect ron spin resonance) , g= 2. 0016 for smoke tar f rom burning coal ; and g = 2. 1008 for the one f rom burning oil. The former was something like the polymer of quinone (Q) / hydroquinone (QH2) ; the latter was similar with the one f rom suspending dust in atmosphere. It was suggested that both the mutagenesis and the carcinogenesis of smoke tar from burning coal may be associated with it s acceleration on the formation of hydroxyl radicals , with the latter′s oxidative damage to DNA , and with property of the free radicals involved.
