三种脂肪酸对人肝癌细胞DNA 合成和甲胎蛋白分泌的影响

THE INFLUENCE OF THE THREE FREE FATTY ACIDS ON THE AFP SECRETION AND DNA SYNTHESIS OF HUMAN HEPATOCARCINOMA CELLS

摘要: 本实验目的探讨具有相同碳原子长度的亚麻酸、亚油酸及油酸处理BEL - 7402 人肝癌细胞后对DNA 合成和甲胎蛋白(AFP) 分泌抑制的影响。放射免疫法检测肝癌细胞分泌甲胎蛋白(AFP) 含量,3H - TdR掺入法测定DNA 合成放射活性(CPM) 。实验结果表明,三种脂肪酸能抑制肝癌细胞DNA 合成,其中亚麻酸、亚油酸抑制作用明显,有显著性差异( P < 0. 01) ,油酸作用较弱( P > 0. 05) ;40 - 50μg/ ml 亚麻酸能明显地抑制肝癌细胞AFP 分泌( P < 0. 05) ,亚油酸和油酸与对照相比无显著性差异( P > 0. 05) 。结果显示,三种脂肪酸分子中含有数量不同的双键,脂肪酸对肝癌细胞DNA 合成和甲胎蛋白分泌的抑制作用存在构效关系,与分子中双键数量有关。本研究扩充了脂肪酸的用途,为癌症病人的保健和治疗提供实验依据。

Abstract: The purpose of this study was to investigate the inhibition effect of DNA synthesis and the AFP secretion on BEL - 7402 human hepatocarcinoma cells in the presence of linolenic acid ,linoleic acid and oleic acid which having the same carbon chains. The AFP concent ration was determinated with radioimmunoassay ,while the DNA synthesis was studied by the 3 H - TdR incorporation (CPM) . The result sindicated that three f ree fatty acids could inhibite the DNA synthesis of human hepatocarcinoma cells. The effect s of linolenic acid and linoleic acid( P < 0. 01) were significant ,which were st ronger than that of oleic acid ( P > 0. 05) . 40 - 50μg/ ml concent ration of linolenic acid could inhibite the AFP secretion of hepatocarcinoma cells obviously compare with the blank ( P < 0. 05) . The same concent ration of linoleic acid and oleic acid had no difference compare with the blank ( P > 0. 05) . The conclusion showed that there was a relationship between the molecular st ructure and the inhibition effect s of three fatty acids containing different numbers of double - bond.
