多沙唑嗪的致突变作用研究

摘要: 应用CHL 细胞染色体畸变试验、Ames 试验和小鼠骨髓嗜多染红细胞微核试验对多沙唑嗪进行了致突变作用研究。结果显示: 312. 5μg/ 皿- 5 000μg/ 皿各剂量组TA97 、TA98 、TA100、TA102 等菌株回复突变率未见明显增加; 513mg/ kg、1025mg/ kg、2 050mg/ kg 各剂量组PCE 微核率分别为1. 7 ‰、215 ‰、216 ‰,与对照组(210) ‰比较无明显差异( P > 0. 05) ;10mg/ ml、20mg/ ml、40mg/ ml、80mg/ ml、160mg/ ml 各剂量组加与不加S9CHL 细胞染色体畸变率均小于5 % ,但发现正常CHL细胞极少见的染色单体易位,应引起重视。

Abstract: Doxazosin , a new antihypertensive agent , was examined for mutagenicity with cultured CHL cell chromosome aberration test , Ames test and micronuclcus test of PCE in mouse bone marrow. The results showed that doxazosin with 5 doses (312. 5ug/ plate - 5 000μg/ plate) didn’t increase the reversion f requencies in st rain TA97 , TA98 , TA100 and TA102 with or without S9 in the Ames test ; It didn’t increase the f requencies of micronucleus in PCE and chromosome aberrations in CHL cells.
