α粒子诱发转化人支气管上皮细胞系BEP2D 中XRCC5 等基因的突变

摘要: 目的与方法:用聚合酶链式反应-单链构象多态性分析(PCR2SSCP) 方法观察α粒子诱发人支气管上皮细胞系BEP2D 细胞转化过程中相关基因的改变。结果:细胞转化过程中,双链断裂修复基因XRCC5 发生碱基突变,改变从转化早期过程就持续 存在;而p16 基因exon2 和hMSH2 基因exon12 未见变化。结论: XRCC5 基因在α粒子照射后早期发生突变,使细胞丧失修复双链断裂损伤的能力,是α粒子诱发支气管上皮细胞转化过程中的启动事件之一。

Abstract: Purpose and Methods : To detect related genes mutation in human bronchial epithelial (BEP2D) cells transformed by α-particles and by using PCR amplification and single2strand conformation polymorphism ( SSCP) . Results : The point mutation of double-strand break repair gene XRCC5 occurred at the early stage of transformation , but no mutation was found in the exon-of p16 gene and exon12 of hMSH2 gene. Conclusion :Mutated XRCC5 gene can't repair DNA lesions induced byα-particles , and plays as one initiator in the process of transformation induced byα-particles.
