高剂量三氯乙烯对L-02肝细胞中Rho GDIα表达的影响

摘要: 背景与目的: 观察高剂量三氯乙烯( trichloroethylene,TCE )对L-02肝细胞中Rho GDIα基因在 mRNA及蛋白水平表达的影响。 材料与方法: 以β-actin为内参,采用蛋白印迹 (western blot) 和实时荧光定量RT- PCR技术对分别40 μmol/L TCE(实验组)和DMSO(溶剂对照组)处理的正常L-02肝细胞中的Rho GDIα蛋白和mRNA水平进行检测。 结果: 高剂量TCE 处理后Rho GDIα mRNA表达量显著下降而蛋白含量明显升高。 结论: Rho GDIα基因表达在高剂量TCE处理后发生了特异性改变,为揭示TCE 对机体的损伤作用机制提供了重要线索。

Abstract: BACKGROUND AND AIM: To study the effect of trichloroethylene(TCE) on the expression change of Rho GDIα gene at both mRNA and protein levels. MATERIALS AND METHODS: Western blot and real time quantitative RT-PCR were employed to detect the protein and mRNA levels of Rho GDIα in L-02 liver cells ,which were treated with DMSO and high concentrations of TCE. β-actin was selected as internal standard. RESULTS: The protein level of Rho GDIα was significantly up-regulated while its mRNA was significantly reduced. CONCLUSION: The Rho GDIα gene expression level showed specific changes after treatment with high concentration of TCE, providing important clues to the mechanism of TCE-induced damage in humans.
