光气致小鼠肺组织细胞凋亡相关基因的表达

Expression of apoptosis_related genes in mouse lungs induced by phosgene

摘要: 目的:研究光气染毒肺水肿小鼠肺组织Bax、c_Fos、c_Jun、Caspase_3 和Fas的表达。 方法: 10只BALB/c小鼠随机分为对照组和光气染毒组,每组5只。正常对照组以空气为对照,染毒组小鼠给予11.9 mg/L的光气,染毒时间为5 min。染毒后4 h,取小鼠肺脏,用4%的多聚甲醛固定。原位杂交和免疫组化测定肺组织Bax、c_Fos、c_Jun,Caspase_3 和Fas的表达。Westen blot 测定Fas蛋白的表达。 结果: 光气染毒组肺水肿小鼠肺组织Bax、c_Fos、c_Jun、Caspase_3和Fas基因的表达较对照组明显增高(P<0.05),且Fas蛋白的表达也升高(P<0.05)。 结论: 光气可能通过Fas/FasL通路诱导肺上皮细胞发生凋亡。

Abstract: OBJECTIVE: To study the expressions of Bax, c_Fos, c_Jun, Caspase_3 and Fas in pulmonary edema induced by phosgene in mice. METHODS: 10 BALB/C mice were divided into control group and phosgene group with 5 mice in each. Mice were exposed to air and phosgene for 5 minutes in control and phosgene group, respectively. Dose of phosgene was 11.9 mg/L. After 4 hours' exposure, all mice were immediately anesthetized. Lungs were fixated by 4% citromint. The expressions of Bax, c_Fos, c_Jun, caspase_3 and Fas gene were determined by hybridization in situ and immunohistochemistry. We also tested the expression of Fas protein by Western blot. RESULTS: Expressions of Bax, c_Fos, c_Jun, caspase_3, Fas gene and Fas protein increased in pulmonary edema induced by phosgene in mice(P<0.05). CONCLUSION: Phosgene could cause induced apoptosis in pulmonary epithelia. It might be regulated by the Fas/FasL pathway.
