叔丁基- 4 羟基茴香醚致突变性研究

摘要: 目的与方法:用Ames 试验、CHL 细胞染色体畸变试验及微核试验对叔丁基- 4 羟基茴香醚(BHA) 进行了致突变性研究。结果:Ames 试验结果在25 - 300μg/ 皿剂量范围内加与不加S9 mix 条件下,4 个菌株的回变菌落数均未有2 倍以上的增加;微核试验显示在171 - 683 mg/ kg. bw 剂量,小鼠骨髓嗜多染红细胞微核率与对照组比较无显著性差异;CHL 细胞染色体畸变试验结果显示在不加S9 mix 条件下10 - 60μg/ ml 剂量范围内染色体畸变率< 5 % ,在加S9 mix 条件下40μg/ ml 剂量组畸变率为7. 5 % ,属可疑阳性,推测叔丁基- 4 羟基茴香醚可能具有致突变性。结论:应对BHA 在食品及食品油中应用的安全性给予重视。

Abstract: Purpose and methods : The mutagenicity of 3 - tert - butyl - 4 - hydroxyanisole (BHA) was examined by Ames test , CHL cell chromosome aberration test in vit ro and micronucleus test . Results : The results showed that BHA with doses 25 - 300μg/ plate didn’t increase the number of reversion mutation in any of the 4 st rains with or without S9 mix in Ames test . The f requencies of micronucleus in PCE were not increased at dose level of 171 - 683 mg/ kg. bw in micronucleus test . In CHL cell chromosome aberration test , the result was suspicious positive because the f requency of chromosome aberration was increased to 7. 5 % at the dose level of 40μg/ ml with S9 mix. Therefore , 3 - tert - butyl - 4 - hydroxyanisole may be mutagenic. Conclusion : We should pay attention to it s application as an anti - oxidant in food and oil product s.
