THE GENERATION OF TRANSGENIC MICE HARBOURING NONSTRUCTURAL 3 GENE OF HEPATITIS C VIRUS

Abstract: To generate t ransgenic mice , the recombinant plasmids inserted the ns3 gene of hepatitis C virus were int roduced into the male pronuclei of mouse fertilizer eggs by microinjection. By using polymerase chain reaction(PCR) and Southern2blot hybridization method we obtained three mice (Founders) and four mice ( F1) harbouring the ns3 gene among 21 off springs born by the pseudopregnant mice and 14 individuals generated f rom the Founders mated with other non2t ransgenic mice , respectively. These t ransgenic mice would be helpful for the development of ns3 t ransgenic mice line and the study for the biological function of NS3 protein in vivo.
