To generate t ransgenic mice , the recombinant plasmids inserted the ns3 gene of hepatitis C virus were int roduced into the male pronuclei of mouse fertilizer eggs by microinjection. By using polymerase chain reaction(PCR) and Southern2blot hybridization method we obtained three mice (Founders) and four mice ( F1) harbouring the ns3 gene among 21 off springs born by the pseudopregnant mice and 14 individuals generated f rom the Founders mated with other non2t ransgenic mice , respectively. These t ransgenic mice would be helpful for the development of ns3 t ransgenic mice line and the study for the biological function of NS3 protein in vivo.

With reverse t ranscription polymerase chain reaction (RT2PCR) and DNA recombinant technique , a full2length cDNA encoding the monooxygenase cytochrome P450 2C9 from human liver was cloned into a plasmid p GEM23Z. The cDNA segment was analyzed by rest riction enzyme mapping and DNA sequencing , and was confirmed to be nothing other than CYP 2C9 cDNA. Then the CYP 2C9 cDNA was subcloned into a mammalian expression vector , ready for development of the cell line expressing CYP 2C9.

Spontaneous mutation rate at hypoxanthine phosphoribosylt ransferase (hprt) locus increased markedly in Hela2MLH1 - cell , mismatch repair (MMR) deficient cell line , it exhibited a 8. 78 and 3. 532fold increase in mutation f requency relative to MMR2proficient HeLa2R9 and HeLa cell line. This phenotype is similar to another mismatch repair defective cell line HeLa2MSH2 - .MNN G tolerance also appeared in HeLa2MLH1 - cell but no MNN G tolerance in HeLa2MSH2 -cell. These data suggest that different MMR gene defect exert s different effect s on mutator phenotypes.

In this research ,sixteen coal samples including seven kinds of coals in Shanxi province was investigated ,which including : bituminous coal stome , coal carbonization ,gas coal ,etc. Each coal sample ext ract was divided into two part s : non2nit rosated sample and nit rosated sample.Ames test ,SOS test ,doublenuiclei micronuclei test , rat bone2marrow cell chromosome aberration test indicated :among sixteen coal ext ract s ,ten were positive in Ames test ,SOS test . One or two or three samples f rom the ten positive est ract s were performed by chromosome aberration test and doublenuclei micronuclei test . Positive rate was 62. 5 %. Test s indicate large concent ration of volitile component , nit rosated ext ract s which are heavier brown liquid were mutagenic. The heavier the color was , the more mutagenic the ext ract s were ,which indicated concent ration of volatile component reflected the organic substance content s indirectly. Large concent ration volatile was in large proportion ,which appeared more mutagenic. This discovery provides a much simpler way to test the dispersible content , which can save large amount of human power and material power.

The mutagenicity of methylene chloride ext racrs of aerosol samples f rom 1995 - 96 in new city region of Kowloon was studied with st rain TA98 and TA100 in Ames Test . It was found that the ext ract samples contained both f rameshift mutagens and base2pair substitute ones. The lowest concent rations produced positive reaction in given monthes were just 5μg/ plate or 2M3/plate , which meant the level of some mutagens in ambient air was higher. The results were also suggested that there were some other mutagens instead of PAH which played an important role in the classical test of air pollution. The mutagenicity of the ext ract s showed obviously a change with the seasons and geographical features(higher in winter and lower in summer) ,as well as a correlation with the change of that in the close region of South China.

The Effects of sperm abnormality in mice treated by three different mutagens , MMS、CP、MMC and with different doses. Based on the analysis of the results , a strategic suggestion was indicated.

Aneuploidy in human population is a serious genetic problem. The major cause of this numerical chromosomal aberration is non - disjunction. Some of the common mutagens such as formaldehyde are known to be able to induce non - disjunction. The purpose of this study is to establish a system for the investigation on the mechanism of non - disjunction and identification of the non - disjunction inducing agent s. Tradescantia ref lex a plant which has high spontaneous frequencies of non - disjunction is an ideal system for the study of aneuploidy induction.

The effect of vitamin A(Vit A) and sodium selenite (Na2SeO3) on rapeseed oil vapor condensate toxicity was studied in mice. The result sare : ①The body weight of rapeseed oil vapor condensate group mice was significantly lower than that of addition Vit A group and Na2SeO3 group ( P < 0. 01) ; ②The heart organ index of rapeseed oil vapor condensate group mice was higher than that of supplyment Vit A group and Na2SeO3 group ( P < 0. 05) ; and ③The rate ofmicronuclei and of sperm aberrations of rapeseed oil vapor condensate group were obviously higher than that of plus Vit A group and Na2SeO3 group ( P < 0. 001 and P < 0. 01) , but rapeseed oil group mice was didn’t ( P > 0. 05) .

The mutagenic and antimutagenic effect s of channel modulating capsule (CMCA) were studied with micromucleus test in bone marrow cells of mice (in vivo) and in the lymphocytes of human blood(in vit ro) . The main result s are as follows ①10 - 50μg/ ml CMCA did not induce micronucleus formation in lymphocytes in vit ro. 10 - 50μg/ ml CMCA inhibited induced micronucleus formation byγ2rays significantly. ②No significant effect of 10 - 50mg/ kg body weight (B.W. ) on MNF in polychromatic erythrocytes (PCE) of mice was found. 10 - 50mg/ kg B. W. CMCA supressed significantly cyclophosophamide (CP)2induce micronucleus formation in PCEs of mice. These result s suggested that CMCA could be used in tumor clinic to reduce side effect s of radio - and chemotherapy.

Polymorphism of metabalic enzyme is one of the factors relevant to the susceptibility to environmental chemicals and heredicary predispositional to carcinogenesis. The polymorphism of N - acetylt ransferase among gast ric cancers is investigated and reported. It was found that 13 among 34 GC patient s (38. 24 %) were slow type of N - AT versus the referring figure 20/ 107 (18. 69 %) in the cont rols. Their gene f requencies were calculated as 0. 6183 and 0. 4323 respec tively , the difference was statistically significant ( P = 0. 022) . The OR of GC for slow N2AT was 2. 69 (1. 16 - 6. 27) , suggesting that slow N2AT should be considered as one possible aetiological factor of GC.

To dectect P53gene mutation and it s mutation protein ,improved aPCR - SSCP silver staining and SP immunohistochemical methods were emploied in 12 borderline epithelial ovarian tumor and 15 benign epithelial ovarian tumor. The result s showed that positive rate of P53 gene mutation was 30. 6 % , and that of the mutation protein was 58. 4 %. The mutation of P53 gene was mainly on it s exon 5 - 8 ,especially on exon 7. In some cases of borderline tumor , P53 gene and it s mutation protein were positive , but nine in benign tumor This indicated that P53gene mutation was a later event in the form ovarian carcinoma. P53gene mutation might serve as a genetic marker to determine the prognosis of ovairan carcinoma. aPCR - SSCP sliver staining method overed the defect of former method ,and improved the accuracy of esamine.
