Study of paclitaxel effects on human lung cancer cell lines with different p53 genotype RUI Meng，CONG Mei，DUAN Yun-you，HOU Chun-mei null    2012, 24 (1): 14-19.   DOI: 10.3969/j.issn.1004-616x.2012.01.004 Abstract （9161）      PDF （654KB）（1564）       Knowledge map    Save OBJECTIVE: To observe the effects of paclitaxel on human lung cancer cell lines with different p53 genotype in vitro. METHODS：A549 (wild-type p53)，H322 (mutant-type p53) and H1299 (absence-type p53) were exposed to paclitaxel at different concentrations (0.1，1.0，10，100，1 000 nmol/L) or different treatment times (4，12，24，48，96 h)，then MTT assay，flow cytometry and western blotting were used to analyze the effects of paclitaxel on cell growth rate，cell cycle progression，apoptosis and the expressions of acetyl-tubulin and p53 protein. At the same time，negative control group (no paclitaxel) and blank control group (only culture medium) were set up. RESULTS：The growth-inhibition effects of paclitaxel on the three human lung cancer cell lines increased along with the response time extension (P＜0.05). Paclitaxel of different concentrations also had different effects on each lung cancer cell line (P＜0.05). The IC50 (50% inhibitory concentration) of paclitaxel for A549 was lowest in the three cell lines，but the difference of chemosensitivity of those cells to paclitaxel was not significant (P＞0.05). For the time-dependent experiment，the induction of apoptosis was time-dependent (P＜0.05) and the percentage of G2/M phase was highest at 12 h when paclitaxel was 10 nmol/L. When treated with 1 000 nmol/L paclitaxel，A549 showed the time-dependent G2/M arrest (P＜0.05)，while H322 and H1299 had the greatest G2/M arrest at 24 h. Prolonging paclitaxel treatments (48 h or more) resulted in appearance of polyploidy cells in H1299. The two concentrations could induce time-dependent apoptosis，but the higher one caused apoptosis later than the lower one. During concentration-dependent experiments， the proportion of apoptosis was highest at 10 nmol/L paclitaxel，while the percentage of G2/M phase increased in parallel with the paclitaxel concentration，when the latter changed from 0.1 nmol/L to 100 nmol/L (P＜0.05). Apoptosis had no relationship with G2/M arrest (P＞0.05). Paclitacxel up-regulated acetyl-tubulin and wild-type p53 protein expressions in time-and concentration-dependent manner，with no influence on mutant-p53 protein. CONCLUSION：Paclitaxel could inhibit the growth of the three human lung cancer cell lines with different p53 status in vitro in time-dependent manner. Different paclitaxel concentrations also had different effects on each lung cancer cell line. Paclitaxel could up-regulate wild-p53 protein expression and induce both p53-dependent and p53-independent apoptosis pathways. p53 genotype of the cells altered the effect of high concentration paclitaxel on cycle arrest，but did not influence paclitaxel chemosensitivity. METHODS：Oral toxicity test (3 administrations within 24 h，total dose 54 g/kg)，Ames test (5 000，1 000，200，40，8 μg/plate)，micronucleus test of bone marrow PCE cell and sperm shape abnormality test in mice (10，5，2.5 g/kg) were used to study the acute toxicity and mutagenicity of Sacha Inchi oil. RESULTS：The oral LD50 of Sacha Inchi oil in mice was more than 54 g/kg. No increase in the number of revertant colonies was found in Ames test. The micronucleus rates and sperm abnormality rates at all doses were not significantly different from the negative control. CONCLUSION：The Sacha Inchi oil when given orally showed no acute toxicity and mutagenicity under our experimental conditions. Related Articles | Metrics

METHODS：Acute oral toxicity test was performed in mice. The genetic toxicity of Res was evaluated by Ames test(Res doses at 5 000，1 000，200，40，8 μg each dish)，bone marrow micronucleus test and sperm shape abnormality test (Res was administered to mice at 7.500，3.750，1.875 g/kg doses). RESULTS：The acute oral maximum-tolerated dose(MTD) of resveratrol was greater than 15.0 g/kg. Ames test，micronucleus test and sperm shape abnormality test showed negative results. MATERIAL AND METHODS: Eighty-one cases of various cervical lesions were analyzed by HPV6/11,HPV16/18 in situ hybridization. Meanwhile, HPV,HSV and CMV were determined in 103 cases of various cervical lesions. RESULTS: The results show that the distribution of positive hybridization signal was consistent with the distribution of koilocytes in HE stain. Of these cervical specimens investigated, the positive rates of HPV 16/18 and HPV6/11 using ISH were 51.1 ％ and 64.7 ％, respectively,the infection rates of HPV 16/18, HPV 6/11,HSV and CMV using PCR were 21 ％,4 ％,23 ％ and 0 ％,respectively. CONCLUSION: The co-operation effect of HPV and HSV might occur in the oncogenesis of human cervical carcinoma. Related Articles | Metrics

MATERIALS AND METHODS: The rate of apoptosis in human breast carcinoma MDA-MB-231 cells was detected by flow cytometry; the activities of Survivin and Caspase-3 proteins were examined by immunocytochemistry and Western blot in untreated and As2O3-treated MDA-MB-231 cells. RESULTS: The percentages of the apoptosis in MDA-MB-231 cells were (28.89±2.47)％,(46.73±3.82)％ and (56.44±4.16)％, in cells treated for 24 hours with 10.0，20.0，40.0 μmol/L As2O3 respectively. As2O3 inhibited the expression of Survivin protein and promoted the expression of Caspase-3 protein. There were obvious dose-effect correlations between As2O3 and its functions. CONCLUSION: The results suggested that As2O3 could induce apoptosis of MDA-MB-231 cells via Survivin protein inhibition and Caspase-3 protein promotion. Related Articles | Metrics

MATERIAL AND METHODS: Young adult NIH mice were treated with a sigle dose of CP(40 or 80 mg/kg) by intraperitoneal injection. The micronucleus freqyuency in bone marrow polychromatic erythrocytes (PCE) at different sampling time 18 , 24 , 48 h after treatment was evaluated. RESULTS: Our data showed: ① The changing trends of the micronucleus frequencies in PCE in the two groups receiving 40 mg/kg and 80 mg/kg of CP were the same at different sampling times. The peak time of the micronucleus incidence was at 24 h after treatment. ② The ratios of PCE to NCE of the two treated groups were not significantly different from the control group. This indicated that the mice bone marrow cells were not suppressed by one CP of 40 or 80 mg/kg by intraperitoneal injection. ③ The micronucleus frequency in PCE of 40 mg/kg was significantly different from that of 80 mg/kg, showing a dose_response relationship. CONCLUSION: CP can be used as a positive control of micronucleus test. The dose range of 40～80 mg/kg was fine, and the optimal sampling time is at 24 h after treatment. Related Articles | Metrics

MATERIAL AND METHODS: With PCR, denaturing high_performance liquid chromatography (DHPLC) and direct sequencing, we screened all exons of the E_cadherin gene for mutations in 80 Chinese gastric cancer patients, 62 of whom had positive family history,we investigated 80 healthy individuals and conducted a case_control study to investigate whether such mutations affect the risk of developing gastric carcinoma. RESULTS: Two of 80 gastric cancer patients carried missense mutation Thr340Ala (heterozygosity). This variation was not found in the control population. We confirmed a T→C polymorphism site at position －13 in upstream of exon 13. The polymorphism was identified in 10 of 80 gastric cancer patients and 12 of 80 healthy individuals and the frequency of this polymorphism did not differ between the familial gastric cancer patients and controls. We found a germline E_cadherin nonsense mutation at codon 751 in exon 14 in 19 of 80 patients analyzed. Only 7 of 80 healthy individuals were Asn751Asn carriers, and significant difference existed between gastric cancer patients and control population, especially between the elder patients and controls. CONCLUSION: Though the germline mutations of CDH1 gene did not occur frequently in familial gastric cancer, CDH1 gene Asn751Asn single nucleotide polymorphism might increase the risk of gastric cancer in Chinese population. METHODS: Sample 70 Pu'er tea randomizely in a Guangzhou tea market. All the samples were assayed with ELISA kit for FB1, DON, T-2 toxin, and with immunoaffinity chromatography - high performance liquid chromatography for AFB1. RESULTS: The AFB1 level of 8 among 70 samples was above the limit of national standard(5 μg/kg), and the DON level of 63 samples above the national standard(1 mg/kg), while the levels of FB1 and T-2 toxin were within the standards(1 mg/kg and 100 μg/kg，respectively). CONCLUSION: AFB1 and DON in our samples were higher and exceeded the allowed standards to some degree. METHODS：SD rats successfully impregnated were divided into 4 groups (15-16 animals per group)，included three Chrysanthemum indicum extract dosage groups (0.68，2.03，6.09 g/kg) and one negative control group. During gestation from 7-16 days，animals were orally gavaged with Chrysanthemum indicum ectract once daily，clinical manifestations and weight of the pregnant rats were collected during the study. On the 20th day pregnant rats were euthanized and the weight of placenta，body weight and length of fetus and the appearance，visceral and skeletal malformations of fetus were examined. RESULTS：No statistically significant difference was found in all the assessed indices(including the weight gain of pregnant rats，average live fetus，absorptive fetus ratio，body weight and length of fetus) between dosage and control groups (P＞0.05). CONCLUSION：Under our experimental conditions，chrysanthemum indicum extract showed no significant effects in the toxicity of SD pregnant rats，had no apparent teratogenicity and embryotoxicity. MATERIALS AND METHODS: Ames test was conducted to determine whether FA and LDXGW ,which contained AA at the concentration of 20 μg/ml, would cause mutagenic changes in the average number of revertants for TA97,TA98,TA100 and TA102. L5178Y/tk(＋/－) －3.7.2C_cells were treated with FA and LDXGE at a concentration of 5 μg/ml by the 96_well microwell method. The plating efficiency (PE), relative total growth (RTG) and the mutant frequency (MF) were determined by the microtiter procedure. RESELTS: FA and LDXGE were considered to be nonmutagenic to Salmonella typhimurium tester strains TA97, TA98, TA100 and TA102 in Ames test. FA was characterized by marked cytotoxicity and mutagenic activity. However LDXGW showed that cytotoxicity and mutagenic activity were largely reduced when made into a compound. CONCLUSION: FA had certain cytotoxicity and mutagenicity while LDXGE could reduce the mutation of tk gene. Related Articles | Metrics

MATERIALS AND MATHODS: Systematic safety evaluation experiments were done using acute toxicity, cellular toxicity and genotoxicity experiments including mice bone marrow micro-nuclear test, Ames test and TK gene mutation experiment. RESULTS: LD50 was higher than 80 g/kg; IC50 in CHO and CHL cells were 55.4 and 19.53 mg/ml, respectively.All toxicity tests were negative. CONCLUSION: It was found that Herba epimedii didn't have mutagenic effects, although it had some toxic effects on the CHO and CHL cells at higher doses. Related Articles | Metrics

METHODS：Experiments included control(only medium)，curcumin only(20 μmol/L)，vincristine only(0.5，1，2 and 4 μg/ml) and curcumin(20 μmol/L) plus vincristine(1 μg/ml). We used cell counting to assess cell growth inhibition，colony formation assay to measure long-term effect of each treatment，DAPI staining to observe cell death type，mitochondrial membrane potential test to evaluate damage on mitochondria，flow cytometry(FCM) to analyse DNA contents and cell cycle distribution，RT-PCR to find responsible genes alterations. RESULTS：Compared with control and individual treatment，after 24 h，curcumin plus vincristine treatment could significantly suppress HepG2 cell growth in cell counting assay (P<0.05)，which was confirmed by colony formation assay. DAPI staining indicated increased apoptosis under combined treatment (P<0.01). Mitochondrial membrane potential was reduced which might explain the increased apoptosis. FCM showed apparent cell arrest in G2/M phase (P<0.05). RT-PCR detected the decrease in expressions of LRP and MDR1 as well as the increase in P21 expression (P<0.05). CONCLUSION：Curcumin plus low concentration vincristine could induce growth inhibition of HepG2，providing a new option for chemotherapy. Related Articles | Metrics

MATERIAL AND METHODS: 72 Wistar rats were randomly divided into six groups including control,VC,VE1,VE2,VE1＋VC and VE2＋VC groups. The trial lasted 8 weeks,and urine and blood samples of each rat were collected at the end of the trial. The activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), level of malondialdehyde (MDA) and DNA damage were assessed. RESULTS: Plasma SOD and GSH-Px activities in VE1 group were 425.21 NU/ml and 440.08 U/ml,respectively which were significantly higher than those of the control group while MDA in this group was significantly lower than that of the control. The same antioxidative enzyme activities in VE1＋VC group were significantly lower than that of the VE1 group. Lymphocyte DNA oxidative damage induced by 10 μmol/L H2O2 in VE1 group was significantly lower than those in other groups and O6-methyl guanine (O6-meG) level in this group was 0.89 mg/g creatinine which was significantly lowered by 50.28％,50.00％ compared with that of control group and that of the VE2 group,respectively. CONCLUSION: Relatively high dose VE could effectively increase antioxidative activity and decrease DNA oxidative and alkyl damages. Using the same dose of VE and VC supplements in combination was found to confer no such advantageous effect. Too high dose VE and VC could decrease the genetic stability. Related Articles | Metrics

METHODS：Micronucleus test and sperm malformation test were performed to analyze the mutagenicity of Henna. RESULTS：The rates of micronuclei in three Henna groups (with concentrations of 5 000，2 500，1 250 mg/kg，respectively) and control group were 0.8‰，1.0‰，0.9‰ and 0.8‰，respectively. There was no significant difference between Henna groups and control group (P>0. 05). The rates of abnormal sperm in three Henna groups and control group were 1.4%，1.6%，1.4% and 1.3%，respectively. METHODS: Kunming mice were randomly divided into the control group, model group, and different concentration SHS groups. The anti-inflammatory effect of SHS was evaluated with croton oil-induced ear edema in mice and the increased levels of inflammatory mediators, nitric oxide (NO). The antioxidant activities were also studied on the effect on malondialdehyde(MDA) levels and enhancement of superoxide dismutase (SOD) activities. RESULTS: SHS produced significant inhibition on croton oil-induced ear edema in mice in a dose-dependent manner. SHS reduced the levels of NO and MDA, and enhanced the activities of SOD. MATERIALS AND METHODS: Testes from 18－22 day old SD rats were removed and decapsulated，then chopped and sequentially digested with two enzymes at 37 ℃：first with 0.25％ trypsin for 20－40 minutes，then with 0.05％ collagenase V for 40－60 minutes．The speed of centrifugation was 800 r/min. The isolated cells were incubated at 35 ℃ in a humidified atmosphere of 5％ CO2．To increase the purity of Sertoli cells，cultured cells were subjected to hypotonic shock(treatment)with 20 mmol Tris_HCl after 48 h of incubation．After 0.25％ trypsin digestion and adherent culture, the cultured cells were identified by HE staining，oil red O staining and transmission electron microscope. The growth curve of Sertoli cells was evaluated as well. RESULTS: Over 95％ of the cultured cells were Sertoli cells．The exponential phase of growth was 4－9 days. CONCLUSION: Highly purified Sertoli cells could be obtained by two step enzymes digestion and hypotonic shock, and identification of Sertoli cells by oil red O was a convenient method. METHODS: Cultured A549 human lung adenocarcinoma cells were exposed to nano- (30 nm) and micro-scale ZnO particles(≤1 μm) at the concentrations of 25, 50 and 100 μg/ml for 6, 12, 24 and 48 h. In the mean time, DMEM culture medium was set up as control. Morphological changes were examined after exposure, methyl tetrazolium cytotoxicity (MTT) assay and the single cell gel electrophoresis(comet assay) were used to determine cytotoxicity and DNA damage. RESULTS: The IC50 of the nano-ZnO and micro-ZnO on the A549 cells were 53.91 and 190.15 μg/ml, respectively. It was found that nano-ZnO inhibited A549 cells significantly at 50 μg/ml, showing a dose- and time- dependent manner. But micro-ZnO inhibited A549 cells significantly only when the concentration up to 100 μg/ml. The slope of the dose-response curve and time-effect curve of the nano-and micro-ZnO was significantly different. In addition, nano-ZnO particles demonstrated a DNA-damaging potential compared to control group in the Comet assay after an exposure of 6 h. Micro-ZnO did not induce any DNA damage. CONCLUSION: Nano- and micro-ZnO particles both demonstrated cytoxicity in A549 cells, but the toxic effects of nano-ZnO occurred earlier in time and lower in dose compared with micro-ZnO. RESULTS: Tumor formation time was found to be (9.6±1.2)d and tumor formation rate was 100％. Within 18 d, tumors was round or round-like shape with distinct borders. Commercially-acquired B16 cells were mostly spindle or irregular whilst B16 cells from tumors were round or round-like or short-spindle. HE staining showed that there were a large number of B16 cells in the transplanted tumors. CONCLUSION: Intradermal tumor model of B16 cells in C57BL mice is helpful to monitor tumor growth in early and middle stages and is one of ideal models in tumor studies. Related Articles | Metrics

METHODS：Mated female rats were randomly segregated into four groups，including three RCT-18 groups (11，37 and 129 mg/kg) and one negative control group (sodium chloride injection)，with more than 25 rats in each group. RCT-18 was administered via subcutaneous injection to timed pregnant rats on gestation days (GD) 6-15 every two days. Clinical signs，body weights，and feed consumption were monitored during the whole gestation. Caesarean section and autopsy were performed on GD20. Uterine content were evaluated for number of implantations，resorptions，live and dead fetuses. The number of corpora lutea in each ovary was also recorded．Live fetuses were examined for gender，body weights，body lengths，tail lengths，and gross external，visceral and skeletal changes. RESULTS：There were no significant differences in maternal and embryo-fetal parameters. MATERIALS AND METHODS： 105 pregnant Wistar rats , 75 for teratogenicity test，30 for micronucleus test, were randomly divided into 5 groups. The rats in experimental groups were treated with 10,20 and 40 mg/kg molybdenum acid ammonium salts, in negative control group received normal saline and in positive group was given 20 mg/ kg cyclophosphamide . Teratogenicity test was scheduled at day 20 of the pregnancy，and micronucleus test at day 14. RESULTS： Compared with negative control group，the increased weight of the pregnant rats , the percentage of live fetuses, the embryonic body weight in 40 mg/kg and positive control groups were decreased(P<0.01) . The micronucleus rate of marrow cells in pregnant rats, the liver blood cells and peripheral blood cells in the fetus, the rate of skeletal and visceral deformities and in 40 mg/ kg and positive control groups were increased (P<0.01) . The embryonic body and tail length in 40 mg/kg and positive control groups were decreased (P<0.05). The rate of external deformity in 40 mg/kg and positive control groups were increased(P<0.05). Percentage of live fetuses in 20 mg/kg group was decreased (P<0.05) . The micronucleus rate of the liver blood cells and peripheral blood cells in the fetus, the rate of skeletal and external deformityies in 20 mg/kg group were increased (P<0.05). The rate of external deformity in 10 mg/kg group was increased (P<0.05). MATERIAL AND METHODS: SD pregnant rats were randomly divided into four groups: Pseudoephedrine 50、100、200 mg/kg and negative control. Conventional teratogenicity test was performed. Female rats was given Pseudoephedrine by gastric intubation at 50,100,200 mg/kg on days 6~15 of gestation. RESULTS: Adjusted weight gain of pregnant rats and body weight of fetuses were comparable to the control at 200 mg/kg. The incidence of dead rats was significantly increased at 200 mg/kg.
