镉金属硫蛋白对小鼠脾淋巴细胞DNA 链损伤和修复的体外实验研究

IN VITRO STUDY ON THE DNA INJURIES AND REPAIR OF SPLENIC LYMPHOCYTE INDUCED BY CADMIUM- METALLOTHIONEIN

摘要: 目的:为研究外源性镉金属硫蛋白(CdMT) 对淋巴细胞DNA 的损伤和修复作用。方法:采用单细胞电泳法(SCGE) 检测DNA 单链断裂(SSBs) ,观察在脱金属硫蛋白(Apo - MT) 和不同终浓度CdMT(0. 1μmol Cd2 + / L 、1. 0μmol Cd2 + / L 、10. 0μmolCd2 + / L 、20. 0μmol Cd2 + / L) 染毒作用30 分钟和60 分钟时DNA 链损伤,和去除受试物后30 分钟、60 分钟、120 分钟时DNA链损伤的修复。结果:DNA - SSBs 尾长在Apo - MT 作用未见增加;在中、高剂量CdMT 染毒30 和60 分钟时都明显增长。损伤后链修复DNA 迁移尾长,在中、高剂量CdMT 组都显著低于细胞染毒时,但明显高于对照和Apo - MT 组。结论:提示CdMT 直接作用可致脾淋巴细胞DNA 链的损伤,同时细胞存在对单链断裂的修复机制。

Abstract: Purpose : To study and compare DNA injuries and repair in BALB/ C mice splenic lymphocyte induced by exogenous cadmium2metallothionein (CdMT) . Methods :Single cell gel elect rophoresis ( SCGE) assay was sed to detect DNA single st rand breaks (DNA2SSBs) when lymphocyte were induced with different concent ration of CdMT for 30 or 60 minutes , and the ability of DNA repair after CdMT was removed for different times from 30 to 120 minutes respectively. Results : It was showed that the comet length of DNA-SSBs did not found change in Apo2MT group , while there were significant increasement of the comet length in mid2 or high-dose CdMT group at both exposure times. And the comet length of each DNA repair times (30 to 120 min) in mid2 or,high-dose CdMT group was significantly shorter than that of DNA injuries , but longer than that of cont rol and Apo2MT group. Conclusion : It is indicated that lymphocyte DNA2SSBs damage would be induced by CdMT exposed directly , and that there were repair activity for DNA2SSBs in spleen lymphocyte in vit ro.
