阿魏酸等五种酚类物质对DNA氧化损伤的影响

摘要: 背景与目的: 用单细胞凝胶电泳法研究阿魏酸、咖啡酸、香草酸、阿司匹林(乙酰水杨酸)和水杨酸五种酚类物质对DNA损伤的影响。材料与方法: 取正常人外周血淋巴细胞与不同浓度(0、50、100、500 μmol/L)阿魏酸、咖啡酸、香草酸、阿司匹林、水杨酸五种酚类化合物于37 ℃下共同孵育30 min后用或不用H2O2于4 ℃下处理10 min,采用单细胞凝胶电泳法对DNA损伤进行分析、比较。 结果: 分别用五种酚类化合物对淋巴细胞进行预处理后,经过H2O2处理,与阳性对照组相比,彗星尾距都有不同程度的缩短,其中阿魏酸、香草酸的作用最明显,并随着剂量的增加,尾距有减小的趋势(各浓度组之间没有显著差异),而咖啡酸、阿司匹林、水杨酸在浓度为500 μmol/L时,尾距较低浓度组(50、100 μmol/L)有所增加,以咖啡酸最为显著。用500 μmol/L咖啡酸、阿司匹林、水杨酸与细胞共同孵育30 min后(未经H2O2处理),咖啡酸可以导致DNA的明显断裂损伤,与对照组之间的差异有统计学意义,而阿司匹林、水杨酸与对照组相比,其彗星尾距没有显著变化。 结论: 上述五种酚类物质均表现出了一定的保护DNA抗氧化活性,其中以咖啡酸保护DNA的作用最弱,而且咖啡酸单独与细胞作用时,可以产生DNA断裂损伤;阿司匹林和水杨酸在高浓度组其尾距较低浓度组又有所缩短,阿魏酸和香草酸抗氧化效果较好。

Abstract: BACKGROUND & AIM: To investigate the in vitro effects of ferulic acid, caffeic acid, vanillic acid, aspirin, salicylic acid on DNA damage using the single cell gel electrophoresis(comet assay). MATERIAL AND METHODS: Lymphocytes from healthy volunteers were pre-incubated with these phenolic compounds at various concentrations(50,100, 500 μmol/L) or 500 μmol/L only, the comet assay was performed on challenged or unchallenged cells in parallel, oxidant challenge being induced by 10 min exposure to 40 μmol/L H2O2. RESULTS: Compared to positive control, the tail moments were all reduced after pretreatment of the cells with different phenolic acids at different concentrations and then challenged with H2O2. The protective effects of ferulic acid and vanillic acid were greater, and the upward trend was shown with the increasing dose. Whereas aspirin, salicylic acid, especially caffeic acid at high dose (500 μmol/L) exhibited greater tail moment than low doses(50 μmol/L, 100 μmol/L). The tail moment showed a marked increment compared to control group after treatment of the cells with caffeic acid (500 μmol/L) at 37 ℃ for 30 min (without H2O2 challenged), whereas aspirin(500 μmol/L) and salicylic acid(500 μmol/L) showed no effects. CONCLUSION: Protective effects were showed by all five phenolic compounds at different concentrations. The protective effects of ferulic acid and vanillic acid increased with dose. However caffeic acid showed less protective effects. In fact, incubating lymphocytes with 500 μmol/L caffeic acid alone actually induced DNA damage.Aspirin and salicylic acid also exhibited better antioxidant effects, but whether they have pro-antioxidant effects deserve further investigations.
