三羟异黄酮对黑色素瘤B16细胞的诱导分化作用

摘要: 背景与目的: 评价三羟异黄酮对黑色素瘤B16细胞的诱导分化作用。 材料与方法: 以4',5,7三羟异黄酮处理黑色素瘤B16细胞1~4 d后,以生长曲线反映其增殖活力,以B16细胞形态、黑色素含量、及流式细胞仪检测细胞周期变化等观察三羟异黄酮对黑色素瘤B16细胞的诱导分化作用。 结果: 用10、30、90 μmol/L的三羟异黄酮作用肿瘤细胞,有不同程度的抑瘤作用。表现为黑色素生成能力增加,细胞生长缓慢。可使B16细胞阻断在S期。 结论: 三羟异黄酮不同剂量(10、30、90 μmol/L)对B16细胞均有明显的分化诱导作用,对体外黑色素瘤增殖有明显的抑制作用。

Abstract: BACKGROUND & AIM: To evaluate the inhibiting effects of Genistein on B16 melanoma cells. MATERIAL AND METHODS: B16 melanoma cells were treated with 10,30,90 μmol/L of Genistein for 1~4 days. Growth curve test served as the index to reflect proliferative activity of B16 melanoma cells.Morphological changes of B16 melanoma cells, melanin content in the supernatant of melanoma cells,distribution of cell cycle were assessed by Flow Cytometery. RESULTS: Genistein at the concentration of 10,30,90 μmol/L inhibited significantly the growth of B16 melanoma cells in a dose dependent manner with statistical significance (P<0.05 vs control). Interestingly, Genistein induced the differentiation of B16 melanoma cells and decreased the melanin granule and number of cells. Flow cytometry showed that numerous cells were arrested at S phase of cell cycle. CONCLUSION: Genistein at the dose of 10,30,90 μmol/L significantly induced differentiation of B16 cells while inhibiting their growth.
