EXPRESSION OF TUMOR NECROSIS FACTOR IN GALLSTONE AND GALLBLADDER CARCINOMA TISSUE

Abstract: Abstract : Purpose : To study the expression of TNF -αmRNA and TNF -αprotein in the development from hyperplasia , dysplasia to carcinoma of gallbladder mucosa and to discuss the pathogenesis of gallbladder mucosa and to discuss the pathogenesis of gallbladder carcinoma. Methods : In situ hybridization and immunohistochemist ry were used to detect the expression of TNF -αmRNA , TNF -αprotein in mucosa of hyperplasia , dysplasia and carcinoma of gallbladder. Results : ①The percentages of positive cells and the numbers of positive cells/per high power field (HPF) which expressed TNF - αmRNA in hyperplasia , dysplasia and carcinoma of gallbladder mucosa cell and mononuclear cells(MNC) were increasing. ②The numbers of carcinoma cells and MNC/ HPF which expressed TNF -αmRNA increased with increasing tumor grades and size. ③The pattern of TNF -αprotein expression was similar to that of TNF -αmRNA , but more f requent and widespread. ④There was a positive linear correlation between tumor cells and MNC of TNF -αmRNA and TNF -αexpression. Conclusion : TNF -αwas involved in the pathogenesis of gallbladder carcinoma induced by gallstone and the TNF -α expression in cancer cells may possibly be a marker for tumor staging.
