To understand the role of mitochondrial DNA(mtDNA) in carcinogenesis , Single2step Method was used to isolate the mtDNA f rom human adenocarcinoma cell line SPC - A - 1. The mtDNA was analyzed by rest riction f ragment length polymorphism(RFL P) with 11 kinds of rest riction endonuclease , which were Pvu Ⅱ,Xho Ⅰ,Pst Ⅰ、EcoR Ⅰ,Bst E Ⅱ,Hind Ⅲ,Hpa Ⅰ,Bcl Ⅰ, EcoR Ⅴ,Sca Ⅰand Xba Ⅰ. Rest riction map of mtDNA f rom SPC - A - 1 cell was obtained by the single and double2digestionn method. It was found that any variation at 32 rest riction2sites failed to be detected in the coding region of mtDNA f rom SPC - A - 1 cell line. But a new site was found at nucleotide 16276( EcoRV) within the noncoding region. These result s indicate that the primary st ructure of gene coding region of mtDNA isolated f rom SPC - A - 1 cell is highly conserved. Whicle the major variation of nucleotide is probably located in the noncoding region.

In this paper , the DNA damage effect s of carbon tet rachlororide , t richloromethane ,dichloromethane , chloroacetic acid and t richloroacetic acid were studied using the single cell gel elect rophoresis technique ( comet assay) in V79 cells cultured in vit ro. The result s showed that all test substances , except trichloroacetic acid , exhibited a clear and reproducible linear dose2dependent increase in DNA migrtion. The DNA breakage capacity and the cytotoxicity of this chemicals have the same order : carbon tet rachlororide >t richloromethane > dichloromethane > chloroacetic acid > t richloroacetic acid. The data also suggested that DNA breakage capacity and the cytotoxicity of the chemicals of chlorinated alky1 were st ronger than that of the kind of chloroacetic acid. Among the chemicals of chlorinated alky1 , the DNA breakage capacity increased with increasing in the number of chlorine substitution.

This report describes a method to separate mRNAs by means of the Polymerase Chain Reaction(Differential Display mRNA) , and identifies the genes related to lung cancer induced by radiation by this method.The RNAs were isolated f rom two pairs of sample , SV40 - immortalized human fetal t racheal fibroblast cell (SHTF) versusαSHTF cell (SHTF cell irradiated byαparticle) , and lung cancer tissue versus normal lung tissue obtained f rom one patient . These RNAs were amplified by RTPCR. The differential expressed gene fragment s were display by autoradiograph or silver nit rate stain. The result showed that the differential display mRNA were established on both cell and tissue samples. The bands stained by silver nit rate is more clear than the bands on X2ray film. The rate of reamplification of differential expressed genes f ragment s stained by silver nit rate is 80 % , higher than autoradiograph , 50 %.

The antimutagenic effect of Cortex acanthopanasia Radicis (CAR) has been studied by mean of micronucleus test of mouse bone marrow polychromatic erythrocyte ( PCE) and mouse spermatid malformation test . The result s indicated that chinese medicine CAR it self has no genotoxic effect . The inhibition rates of CAR (1g/ kg ,2g/ kg ,4g/ kg) to micronuclei was respectively 73. 27 % ,50. 5 % and 63. 37 % ( P < 0. 01) ,and inhibition rates of CAR to spermatid malformation was 84 % (1g/ kg) , 81. 45 %(2g/ kg) and 73. 45 % (4g/ kg) ( P <0. 01) . It is suggested that CAR can significantly inhibit the genetic damage of germ cells and stomatic cells induced by MMC.

In this paper, a new method to assess the environmental safety of pesticide has been proposed in terms of sperm deformity of the earthworm ( Eisenia f oetida) . The result s showed that the main sperm morphological deformity appeared to be shape changes of the sperm heads. Significant induction of sperm deformity ( P< 0. 01) and dose2effect relationship displayed when the earthworms were exposed to imidacloprid at the concent rations of more than 0. 5mg/ kg dry soil. However , The sperm deformity f requency of RH - 5849 - exposed groups did not show significant difference ( P < 0. 05) f rom the negative cont rol until the dose reached 100 mg/kg dry soil. This study suggest s that the detection of sperm abnormality in earthworms could provide an early warning system of polluted environment .

Before and after the operation (1992 - 1996) of Daya Bay Nuclear Power Station , the apoptosis in amphibian animal cells were oberserved and compared in this paper. Result s showed that there were no significant changes among the morphological markers of apoptosis and suggested that the operation on the nuclear power station during the first three years had no significant effect s on the apoptosis of amphibian animal living nearby the station.

We analysed cytogenetics indexes from 29 women and 27 women who had been implanting NorplantR Ⅰand CLa as a long2term cont raceptive system , respectively and f rom 19 women as the cont rol group. The results suggested that NorplantR Ⅰmay be induce the existence of an increase mutagenic environment in cells of its impanters and significantly higher f requencies of chromosome aberration and sister chromatid exchange , the problems of NorplantR Ⅰsubdermal implant for long term cont raception should be attached importance to and that this kind of CLa could not be a clastogen or mutagen2carcinogen.

The aim is immortalizing the human fetal t racheal fibroblast cells to study the cell neoplastic transformation. The primary human fetal t racheal fibroblast cell was t ransfected with pZIPSV40T vector. The colonies that resistant to G418 were collected , and passaged to immortalize. The biological characters immortalized cell had been determined. Result : the immortalized cell morphology is shorter than normal , arrange un2regular. The cells grow well at p50. Southern blot confirmed the presence of T antigen as integrated form in immortalized cell DNA. The index of growth period was fourth day after plated. All the cells could not form colony in soft agar.No tumors formed after SC injection of the cells in nude mice. The result confirmed that human fetal t racheal fibroblast cell can be immortalized by SV40T gene.

To look for an ideal chemical for surface modification of asbestos , the effect s of aluminium cit rate ,mixed rare earths and sodium selenite on the induction of chromosomal aberrations and sister chromatid exchanges (SCEs) by chrysotile in human embryo lung cells were observed. The result s showed that chrysotile caused increases of chromosomal aberrations and SCEs in HEL cells , but the three compounds - pret reated chrysotile induced less chromosomal aberrations and SCEs than the native chrysotile. These result s indicated that the pret reatment of chrysotile with three compounds can inhibite the damage effect s of chrysotile on the chromosome of HEL cells and may reduce the carcinogenicity of chrysotile.

The mice bone2marrow polychromatic erythrocytes (PCE) micronucleus test in vivo to check the antimutagenic effect of fortified full2nut ritional milk (FFNM) on mutagenicity induced by cyclophosphamide (CP) was carried out and anticarcinogenic potentials of FFNM on t ransplanted sarcoma S180 , bladder carcinoma and Ehrlich ascites carcinoma in mice were tested. The experimental result s indicated that FFNM with preventive administ ration significantly inhibited CP induced bone2marrow micronucleus formation in mice , which suggest that FFNM exhibited a protective effect on mutageninduced chromosomal breakage , i. e. it have a antimuta genic action. In addition , to a certain degree , FFNM might inhibit growth of t ransplanted substantive tumor.These result s suggested that FFNM might be of potential value in it s clinical applications.

The antimutagenicity of A - CM marine biological preperation was studied with several genotoxicological test s which included antigenetic reverse mutation test in S almonella typhimurium st rain TA 100 , the test of antimicronucleus effect in vivo mice bone marrow on PCEs induced by common mutagens Cd2 + and NO -2 ,and DNA damage resistance test . Result s consistently demonst rate that A - CM preperation has significant antimutagenic effect on gene mutation and chromosome clastogen in all above tests.

The reproductive toxicity of vindesine sulfate (VDS) in Wistar rat s was carried out . Before mating trial , VDS was iv given once a week at 0. 05 , 0. 1 and 0. 2 mg/ kg to male rat s (9 doses) and to female rats (3doses) . The administ ration was same to both rat s during the mating t rial (1∶1 cohabitation , for up to 2 weeks)and to mated females on day 6 and 12 postcoitus (pc) . Mated females were killed on day 20 pc. As compared to the saline cont rol , the body weight gain of males f rom every dosage group is less. In high dosage of VDS ( 0. 2mg/ kg) , the percentage of dead fetus is higher and the tail length and body weight of survival fetuses is less.The percentage of delayed skeletogency of every dosage group is higher. No malformation of survival fetuses was observed in every group. The study indicated that VDS is embryotoxic but not teratogenic in Wistar rats and has no obvious effect on reproductive performance of both sexes.

Yi Tong Shu, a Chinese medicine, was examined for mutagenicity with cultured CHL cell chromosome aberration test , Ames test and micronucleus test of PCE in mouse bone marrow. The result s showed that Yi Tong Shu with 5 doses (0. 00625ml/ plate - 0. 1ml/ plate) didn’t increase the reversion mutation frequencies in st rain TA97 , TA98 , TA100 and TA102 with or without S9 in the Ames test ; It didn’t increase the frequencies of micronucleus in PCE and chromosome aberrations in CHL cells.

The mutagenicity of tea green pigment was studied. No mutagenic proof s were found in Ames test . The f requencies of micronucleus in bone marrow PCE cells of mice t reated with tea green pigment at the doses of 800mg/ kg ( 1/ 16LD50) 、1 600mg/ kg ( 1/ 8LD50) 、3 200mg/ kg ( 1/ 4LD50) showed no significant difference when compared with that of cont rol. The distinctly increasing separation of sexual chromosome was found in mice t reated with tea green pigment at the 3 200mg/ kg (1/ 4LD50) and 800mg/ kg (1/ 16LD50) groups in primary spermatocyte chromosome aberration assay. However , no signicant difference in f requencides of chromosome aberration and separation of ordinary chromosome in primary spermatocyte was observed between the t reated groups and the cont rol.

Doxazosin , a new antihypertensive agent , was examined for mutagenicity with cultured CHL cell chromosome aberration test , Ames test and micronuclcus test of PCE in mouse bone marrow. The results showed that doxazosin with 5 doses (312. 5ug/ plate - 5 000μg/ plate) didn’t increase the reversion f requencies in st rain TA97 , TA98 , TA100 and TA102 with or without S9 in the Ames test ; It didn’t increase the f requencies of micronucleus in PCE and chromosome aberrations in CHL cells.
