The antimutagenicity of 9 kinds of chemical compounds (osthol , Bergaten , Isopimpinellin , Imperatorin , Xanthotoxol , Xnthotoxin , Thymine , Uracil and unknown one that need authenticating) was studied with the Ames test and the micronucleus and chromosomal aberretion assays in mice in vivo. These compounds were isolated f rom the water ext ract of Cnidium monnieri (L. ) Cuss in Chinese t raditional medicine. The results showed that osthol. , bergapten , isopimpinellin , imperatorin , xanthotoxol and xanthotoxin had inhibited activity of mutagenicity of aflatoxin B1 . And osthol , bergapten , isopimpinellin , imperatorin also had high activity in inhibiting the chromosomal aberrations and micronuclei that induced by cyclophosphamid in vivo. The other three compounds ( Thymine , Uracil and unknown one that need authenticating) showed no antimutagenicity.

Sperm chromosome of subject s with hepatitis B virus infection were prepared using interspecific in vit ro fertilization with zona2f ree hamster eggs. Fluorescence in situ hybridization ( FISH) to sperm chromosome spreads was carried out with biotin2labelled full length HBV DNA. There was significant difference in the frequencies of sperms with chromosome aberrations between subject s with HBV infection and cont rol. FISH showed that HBV DNA integrated into sperm chromosomes , and that multisite and nonspecific integrations were the features of HBV in sperm chromosomes. These result s indicate that HBV infection affect s on the sperm chromosome broadly.

To understand the mechanism of mutagenesis inhibitory effect of Turmeric oil , the authors designed a series of testing procedures to examine the inhibitory effect by Turmeric oil on the mutagenesis induced by Mitomycine (MMC) in Escherichia Coil lysogenic st rain of GY5027 as indicated by GY4015 as following ways : A.GY5027 was added after Turmeric oil and MMC had been put together for 20 minutes. B. Turmeric oil was added after GY5027 had been t reated with MMC for 20 minutes. C. MMC was added after GY5027 had been treated with Turmeric oil for 20 minutes. D. GY5027 , Turmeric oil and MMC were put into together at the same time. Result s found that in all of the above A , B , C and D experimental designs , the mutagnesis was significantly inhibited by Turmeric oil. This indicates that Turmeric oil might be able to inhibit mutagenesis both intercellularly and ext racellularly and repair genetic damages caused by genotoxicant .

To study the f requency of ApoB、ApoE alleles in population of Southern Zhejiang , we collected 60 random healthy individuals to detect the polymorphic sites EcoR Ⅰ、Xba Ⅰof ApoB. Hha Ⅰof ApoE by PCR-RFL Ps. The result showed X- allele and E+ allele of Xba Ⅰand EcoR Ⅰ rest riction sites of ApoB have an advantage in f requencies of alleles having relative f requency of 0. 983 and 0. 967 , respectively. X+ and E- alleles were very rare. The f requencies of the three ApoE alleles were E2 = 0. 02 , E3 = 0. 86 , E4 = 0. 12.

To set up the animal model of precancerous lesion of rat’s pancreatic acinar cellular cancer , learn its biological feature and K2ras gene code 12 mutation. Male Wistar rat s were injected Azaserine 30mg/ kg (ip) . 4 months later , rat s’pancreas were autopsied and K2ras gene code 12 mutation were detected by PCR2SSCP. The incidence of precancreous lesion of rat s’pancreatic acinar cellular cancer was 42. 1 %. Azaserine could induce precancerous lesion of rat s’pancreatic acinar cancer successfully , K2ras gene code 12 mutation was early change during this process.

For the purpose of elucidating the genotoxic dose and genotoxicity characteristics of selenium , healthy human peripheral blood lymphocytes (PBL) in vit ro were t reated for 72h by supplementing with different doses of sodium selenite , then the changes of the f requrncies of sister2chromatid exchanges (SCE) and chromosome aberrations were observed. The result s showed that when the culture of PBL was supplemented with sodium selenite at 0. 05mg/ L and 0. 25mg/ L , the f requencies of SCE and chromosome aberrations were not significantly increased ( P > 0. 5 and P > 0. 3 respectively) , when at 0. 75mg/ L and 1. 50mg/ L , the f requencies of SCE and chromosome aberration were increased obviously ( P < 0. 01 and P < 0. 001 respectively) . High doses of sodium selenite could cause chromosome deform and cent romere segregate premat ruely. It is suggested that high dose selenium possesses mutagenic effect and might possess many other cytotoxinic effects.

The effect of vitamin E (Vit E) on the rate of micronucleus and of sperm deformity in mice were observed with the micronucleus test and the sperm deformity analysis. The experimental data were calculated by statistical U test . The result s showed the rate of micronucleus and of sperm deformity of mice t reated with salad oil vapor condensate at 5ml/ kg and 10ml/ kg groups were abviously higher than that at 2. 5ml/ kg and cont rol groups ( P < 0. 01) . After the mice were supplied Vit E at 20 mg/ kg , the rate of micronucleus and of sperm deformity were significantly lower than that of salad oil vapor condensate group ( P < 0. 01) . The inhibitory effect of Vit E on the toxicity of salad oil vapor condensate was significant in mice.

The mutagenicity of Ethrel was studied by Ames test , sperm deformity test and marrow micronucleus test in mice. The result s showed that Ames test was negative , but the rates of marrow cells micronucleus and spermatic deformity in high dosage group were increased and statistically significant difference was observed between t reated and cont rol groups. It is indicated that Ethrel has potential mutagenicity for somatic cells and reproductive cells in mice.

The genetic toxicity of tet randrine on mice was studied by sperm deformity test ,marrow micronucleus test and sister chromatid exchange. The results showed that the rate of spermatic deformity and marrow cells micronucleus in mouse was increased significantly in growps t reated with tet randrine , and there was dose-response relationship , but the change of the f requencies of sister chromatid exchanges was not obvious. It is concluded that tet randrine has genetic toxicity to experimental animal.

This article is about the study on genetical toxical effect s of five home2made edible synthetic pigments by suing bioassay. Result s showed that the synthetic pigment s with certain dose range would evidently induce the increase of micronucleus frequencies of Tradescantia Pal udosa tet rads , The micronucleus frequencies reached significant or highly significant difference as compared with cont rol ( P < 0. 05 , P < 0. 01) , There was also the obvious dose2response relationshop. F test also showed that significant difference of tet rad micronucleus frequencies induced by five pigment s. Meanwhile they could induce the increase of micronucleus f requencies of Vicia f aba root2tip cells , the test indicated an ultimate difference between higher dose groups and the cont rol group. The study suggested that the edible synthetic pigment s could induce chromosomes damage in plant sex cells and somatic cells , and that had genetical toxical effect s with tested dose.

Mineral water with high radiation was tested to evaluate it s genetic toxicity. In the experiment the Kunming mice were randomly divided into 3 groups , one group , fed with tap water and the other two with different highly2radiated mineral water. Samples were collected on the 50th day ,100th day ,150th day , respectively. Result s showed that the f requencies of the micronucleus and number of motile sperms of the mice drinking highly-radiated mineral water are significantly different ( P < 0. 05) as compared with those drinking tap water at certain period of time. Thus it suggested that the mineral water with high radiation might possess potential mutagenic effect on the animals.

The analyses of micronuclei in peripheral blood lymphocyte were performed in 40 female workers of printing house. The result s indicated that the rates of positive micronucleus , micronucleus cells and micronuclei in the printing workers were much higher than that in cont rol ( P < 0. 01) . The f requencies of micronucleus cells and micronuclei obviously increased as length of service goes on. The possible cause that the micronuclei increased in printing female workers was discussed.

Inhibitory effect of L aminaria japonica polysacchrides (L PS) on micronucleus rate induced by cyclophosphamide (CP) had been studied by mean of micronucleus test in mouse bonemarrow polychromatic erythrocyte (PCE) . Micronucleus rates of positive cont rol group (CP 40mg/ kg. bw) , and three L PS tested groups (L PS 500mg/ kg , 1000mg/ kg , 2000mg/ kg) was 28 ‰, 22 ‰, 12 ‰, 11 ‰, respectively. There were significant differences tested groups (1000mg/ kg , 2000mg/ kg) as compared with positive cont rol group. The results showed that L PS possessed inhibitory effect s on mutagenesis indued by cyclophamide.
