纺锤体毒剂和断裂剂体外诱发V79 细胞多核与微核效应的差异

摘要: 采用长春新碱、秋水仙素(纺锤体毒剂) 、丝裂霉素C 和环磷酰胺(断裂剂) 对V79 细胞染毒,发现两类诱变剂在诱发多核与微核的敏感性和强度上存在显著差异。当剂量较高时,四种诱变剂都可诱发多核与微核,但纺锤体毒剂以诱发多核为主,多核细胞率与微核细胞率的比值为2. 0 - 3. 5 ;而断裂剂以诱发微核为主,此比值倒置(0. 08 - 0. 28) 。当剂量低至一定程度时,纺锤体毒剂只诱发多核(不诱发微核) ,而断裂剂则只诱发微核。还发现两类诱变剂间此比值的差异远大于微核面积或直径的差异。本研究结果提示在体外微核试验中同时计数微核细胞率和多核细胞率可能对鉴别纺锤体毒剂和断裂剂有意义。

Abstract: Two aneugens : vinblastine (Vin) and colchicine (Col) ,and two clastogens : mitomycin C (MMC) and cyclophosphamide (CP) , were applied to Chinese hamster lung (V79) cells respectively in vit ro for micronucleus test . The result s showed that the increases of incidence of multinucleated cells ( Imu) and that of micronucleated cells ( Imi ) in response to aneugens and clastogens were in different sensitivities. At relatively low concent rations , Vin and Col induced only Imu (but not Imi) , while MMC induced only Imi ; At higher concent rations , both Imu and Imi increased in a concent ration dependent manner , however , Vin and Col induced multinuclei predominantly with the ratios of Imu to Imi 2. 0 - 3. 5 ,while MMC and CP induced micronuclei mainly , with Imu/ Imi of 0. 08 - 0. 28. This intensity of difference in Imu/ Imi was much greater than that in areas of micronuclei between aneugens and clastogens. The result s indicate that in micronucleus test in vit ro , observation of both Imu and Imi might be valuable for distinguishing aneugens f rom clastogens.
