应用蚕豆根尖微核技术和彗星试验监测扬中地表水遗传毒物污染的研究

摘要: 目的与方法:本文应用蚕豆根尖微核技术和彗星试验对扬中市不同水体的遗传毒物污染状况进行了初步调查。结果:结果表明,扬中市的遗传毒物污染集中在汇集了乡镇工业废水和生活污水的排污河道和死水沟塘,各主要河道及通江闸口基本没有污染。对其中的新坝大桥(基本无污染) 、联合粮管所塘(中轻度污染) 、扬子河、红星河(严重污染) 水样的有机提取物进行人外周血淋巴细胞的彗星试验,以检测其引起的DNA 损伤。结果表明扬子河、红星河水样引起的DNA 损伤最为严重,其次为联合粮管所塘水样,新坝大桥水样最轻,趋势与蚕豆根尖微核试验结果一致。然而彗星试验结果显示四个样点的有机提取物引起的DNA 损伤与阴性对照相比均有显著差异( P < 0. 01 ) ,结论:上述结果表明彗星试验在检测遗传毒物污染方面比蚕豆根尖微核试验更为敏感。

Abstract: Purpose and Methods : In this study , the micronucleus test and comet assay of V ice f aber were used to screen pollution situation of the surface water of Yangzhong city. Results : The result s showed that the water of Yangzi river and Hongxin river was heavily polluted (Pollution Index > 3) and pollution of other water body was slight . Using the comet assay to detect the DNA damage induced by the organic ext ract s f rom four water samples , the result s indicated that both the comet assay and the micronucleus test were able to detect the geno2 toxic potential of different water body and the comet assay seemed to be more sensitive than micronucleus test . Conclusion : Combination of the comet assay and the micronucleus test might therefore be recommended for monitoring the genotoxicant pollution of water body.
