In this research Fluorescence in situ hybridization ( FISH) and antikinetochore antibodies (CREST Syndrome serum) were used in determining the effect of inducing aneuploid of GMA. The two techniques mainly detected the proportion of cent ric signals in micronuclei induced by GMA. The result s showed that FISH positive ratio of GMA was 40. 9 - 51. 5 % , but for CREST2staining it was only 17. 0 - 24. 1 %. It indicated that GMA was a suspected spindle poison and could damage the cent ric protein. The metaphase analysis demonst rated that GMA had significantly clastogenic effect , which mostly happened in cent romere region.

The delayed expression of chromosomal instability could be induced by 137 Cs - γ ray in mouse marrow stem cells in vivo and in vit ro. The result s showed that 1) , After 14 days incubating of irradiated BALB/ C mouse marrow stem cells , the unstable chromosomal aberration still appeared in CFU - A. 2) , The appearance of the unstable chromosomal aberration could persist for above 30 days in irradiated A4 cells. The statistical analysis of dose2effect response was obversed. 3) , The chromosomal instabiliy could be t ransmitted in BALB/ C mice in vivo for up to 30 days.The f requencies for chromosomal aberrations on 30th day is higher than that of 5th、10th and 20th day. The effect s of chromosomal instability t ransmission is unkowns. It is possible that the delay of chromosomal instability expression lead to cells death and cells t ransformation.

The effect of diethylstilbest rol on urethan2induced mouse lung tumorigenesis was assessed by a single int raperitoneal injecting of urethan (500mg/ kg) or/ and multi int ramuscular injecting of diethylstilbest rol (5mg/ kg or 50mg/ kg) . All mice were sacrificed 18 weeks after administ ration , and the lung tumors were examined histopathologically. Diethylstilbest rol did not produce an elevated lung tumor response when administered alone , but it produced a statistically significant enhancement of incidence of tumors , average number of tumors , incidence of cancers and constituent ratio of malignant tumors when given in conjunction with urethan. The resul s indicated that diethylstildest rol may be a promoter in lung tumor formation.

The genotoxicities of Deoxycholic acid and Lithocholtic acid were tested by Ames test and chromosome aberration test in vit ro. The malignent transformation effect of Deoxycholic acid and Lithocholic acid on NIH/ 3T3 cells was observed , The inhibitory effect of Deoxycholic acid and Lithocholic acid on gap junction intercellular communication ( GJ IC) in NIH/ 3T3 cells was investigated with Scrap Loading/ Dye Transfer test (SLDT) . The result s indicated that neither Deoxycholic acid nor Lithocholic acid has gene mutation , chromosome aberration and cell malignant t ransformation in vit ro , Therefore , it did not demonst rate that bile acid is a genotoxic carcinogen. But result showed that both Deoxycholic acid and Lithocholic acid could inhibit NIH/ 3T3 cells GJ IC obviously. It means that bile acid may play a role as promoter in carcinogenesis.

Vanadium is a proven animal teratogen which can induce dilation of latermal vent ricles and skeletal abnormal in rat , although the mechanism of it s developmental toxicity is unknown. In this paper , the micromass rat embryo midbrain cell culture was used in a affort to determine potential mechanisms by which Vanadium Pentoxide (V2O5) may exert it s teratogenic effect . In vit ro test , a dose2dependent inhibition of differentiation and proliferation of midbrain cells were observed. The 50 % - inhibitory concent ration ( IC50) for cell differntiation and cell survial or proliferation 50 % the cont rol value ( IC50) are 6. 8 and 9. 1μmol/ L , respcectively. In vivo/ in vit ro test , V2O5 could inhibit cell differentiation at 2. 0 mg/ kg body weight , but inhibit cell surival or proliferation at 5. 0mg/ kg. According to class standard of Renault , el al (1989) , V2O5 may be a specific inhibitor of embryonic midbrain cell.

The purpose of this study was to observe the antitumor effect and toxic and side effect of lithium carbonate (Li2CO3) on cyclophosphamide (CP) . All mice were inoculated tumors (hepatocarcinoma H22 or sarcoma S180) according to the standard method. Li2CO3 20 or 40 mg/(kg. d) was administered for 10 da ys. CP (90 or 180 mg/ kg) was administered once by ip. It was shown that combinated application of Li2CO3 and CP significantly enhanced the inhibition rates of tumors (H22 , S180) in mice , and remarkably increased the number of white blood cell (WBC) of the tumor2bearing mice (H22 , S180) , and remarkably decreased the f requency of micronucleus (MNF) of bone marrow polychromatic erythrocyte (PCE) of the tumor2bearing mice. These result s indicated that Li2CO3 could enhance significantly the effect s of CP to inhibite tumor in mice , and remarkably reduce the toxic and side responses of decreasing WBC and increasing MNF caused by CP.

Both the organic and aqueous ext ract s of smoke tar f rom burning coal could accelerate hydroxylative degradation of deoxyribose (dR) of deoxyadenosine (dA) in Uderf riend reaction system , and showed dose2effect relationship to some extent . However , smoke tar f rom burning oil showed none of these effect s. Free radicals were determined by ESR(elect ron spin resonance) , g= 2. 0016 for smoke tar f rom burning coal ; and g = 2. 1008 for the one f rom burning oil. The former was something like the polymer of quinone (Q) / hydroquinone (QH2) ; the latter was similar with the one f rom suspending dust in atmosphere. It was suggested that both the mutagenesis and the carcinogenesis of smoke tar from burning coal may be associated with it s acceleration on the formation of hydroxyl radicals , with the latter′s oxidative damage to DNA , and with property of the free radicals involved.

Male SD rat s were fed orally with Ge - 132 100 , 250 and 500mg/ kg each day for 25days , Cyt P450 of liver cell of high dose group was inhibited significantly ( P < 0. 05) , EROD activity was also inhibited about 50 percent in the group compared with the cont rol. The animals were pret reated with Ge - 132 500mg/ kg/ d for 20 days , then give PB and 3 - MC , found that the inhibition of P450 induced by PB was not obvious , but there was 33 percent inhibition of EROD activity induced by 3 - MC.

In this paper , the mutation and teratogenic effect of the OF23 were investigated. The LD50 was determined as 3. 3g/ kg in mice by oral. The result s indicated that the micronucleus rate was not significant difference ( P > 0. 05) in dosage of 0. 17 - 1. 65/ kg/ day and the marrow chromosome aberration was negative in dosage of 0. 03 - 0. 17g/ kg ,but in teratogenic test , the weight of the female rat s and its fetus were lighten with the dosage increasing , this result shows the OF23 have toxic effect on fetus in long2term administ ration.

The micronucleus assay in mouse marrow PCE and chromosome aberration assay in mouse marrow cells have been used to investigate the inhibitory effect of flesh mulberry juice on mutation rate that was enhanced by cyclophosphamide. The result s showed that the flesh mulberry juice can significantly reduced the PCE micronucleus rate and chromosome aberration rate that was enhanced by the cyclophosphamide in mouse marrow ( P < 0. 05 or P < 0. 01) . It also showed that these effect s have well dose - effect dependent relationship. The relative coefficient is - 0. 94 and - 0. 98.

This paper reported the result s of mutagenesis of sodium ozagrel OKY- 046 assa yed with the Salmonella mutagenicity test Ames test , micronucleus test of polychromatic erythroblast on mouse bone marrow and aberration of sperm test . The result s showed that f rom 0. 05 -500μg/ plate , OKY - 046 didn’t reduce the reversion f requencies in st rain TA98 , TA100 and TA102 ; OKY- 046 couldn’t increase the f requency of micronucleus at the doses of 306. 2mg/kg , 612. 4mg/ kg and 1224. 8mg/ kg ;OKY- 046 didn’t also increase the aberration rate of sperm at the doses of 245mg/ kg , 490mg/ kg and 980mg/ kg , therefore , it is believed that OKY - 046 may be safe on potential mutagenesis.
