Purpose : To develop a inducible system for expression of Trail in E. coli . Methods : RT2PCR and sequencing were used to clone and conform Trail gene. The production of expression was analyzed by SDS2page.Results : Trail cDNA was cloned by RT2PCR. After sequencing , we const ructed the inducible vector for expression of Trail in E. coli . The vector was t ransformed with TG1. Trail was expressed by induced of changing temperature f rom 30 ℃to 42 ℃. The production was conformed by SDS2page. Conclusion : This study will provide new insight into physiological and pathological roles of Trail2mediated cytotoxicity and it s potential application to tumor immunotherapy.

Purpose and Methods : It’s necessary to const ruct the conserved amino acid residue mutations of HDAC1 for studing the functions of the coserved amino acid residues of HDAC1. On the base of cloning of HDAC1 cDNA , Three mutant s of HDAC1 , which contain the conserved three amino acid residue mutations respectively were made by the Altered Site Ⅱin2vit ro mutation system , and confirmed by DNA sequencing. Results and Conclusion : The result s were that C151A , Y188F , S197A mutant s of HDAC1 were obtained , and made the base for further studing the function affection of conserved amino acid residue mutations.

Purpose : To study and compare DNA injuries and repair in BALB/ C mice splenic lymphocyte induced by exogenous cadmium2metallothionein (CdMT) . Methods :Single cell gel elect rophoresis ( SCGE) assay was sed to detect DNA single st rand breaks (DNA2SSBs) when lymphocyte were induced with different concent ration of CdMT for 30 or 60 minutes , and the ability of DNA repair after CdMT was removed for different times from 30 to 120 minutes respectively. Results : It was showed that the comet length of DNA-SSBs did not found change in Apo2MT group , while there were significant increasement of the comet length in mid2 or high-dose CdMT group at both exposure times. And the comet length of each DNA repair times (30 to 120 min) in mid2 or,high-dose CdMT group was significantly shorter than that of DNA injuries , but longer than that of cont rol and Apo2MT group. Conclusion : It is indicated that lymphocyte DNA2SSBs damage would be induced by CdMT exposed directly , and that there were repair activity for DNA2SSBs in spleen lymphocyte in vit ro.

Purpose and Methods :Using liquid scintillation count assay and  I2postlabeling assay , DNA spontaneous synthesis、DNA2protein crosslinks (DPC) and unscheduled DNA synthesis (UDS) were detected to study DNA synthesis、repair function and DNA damage condition in human peripheral blood cells of arsenism caused by burning coal. Results : The DNA synthesis of non-patient s and patient s in local area obvious decreased , while DPC level increased with the development of poisoning , and UDS increased only in the patient s , the difference were all significant . Conclusion : It suggested that arsenic containing in coal might induce DPC in early stage and severely hurt DNA ; and it might induce UDS , the DNA synthesis and repair ability were seriously rest rained. It might be one of the reasons causing dermal cancer.

Purpose and Methods : This paper observed the effect s of Vitamin E Succinate (VES) on cell growth ,DNA synthesis and the appearance of apoptosic morphological change in human gast ric cancer SGC - 7901 cell line in vit ro. Results : The result s showed that VES (5μg/ ml 、10μg/ ml and 20μg/ ml) could obviously inhibit SGC27901 cell growth and DNA synthesis in a dose-dependent manner , and induce apoptosis of SGC27901 cell.Conclusion : These result s indicate that the inhibition of VES on SGC27901 cell growth was by the way of inhibiting cellular DNA synthesis and inducing apoptosis.

Purpose and Methods : Using immunohisthocemical assay , the expression of cyclin D1 protein of lithium carbonate on the development of rat hepatic carcinoma induced by aflatoxin B1 and it s significance were studied. Results : The expression of cyclin D1 occurred in the early stage (the 6th week) and significantly increased in the 10th week. The positive rate of positive cont rol group (B) was the highest and that of group t reated with Li2CO3 simultaneously (C) and group pre2t reated with Li2CO3 . (D) significantly decreased , but group C > group D. Conclusion : It suggested that Li2CO3 has the effect on anti2expression of cyclin D1 in aflatoxin B1 induced rat hepatic carcinoma , and can rest rain cell continuous roliferation , so it may be anticarcinogenic ; and that the immunohistochemical assay of cyclin D1 may help early diagnosis and monitoring.

Purpose : Measuring the amounts of fluorescence by bivariate flow cytomet ry ( FCM) , we examined early stage of apoptosis in 6 vicious gast ric tissues and corresponding normal ones with a double staining method of Annexin V(AV) and propidiom iodide (PI) . Methods : From 6 vicious and 6 normal gast ric tissues of 1cm3 ,single2cell suspension were preparated and stained with fluorescein isothiocyanate ( FITC)2labelled AV and PI ,and AV binding was assessed using bivariate FCM. Results : The separated cells incubated with AV and PI enabled three distinct populations of cells(apoptotic , necrotic and living cells) to be identified and sorted by FCM.Comparing normal and vicious tissues , differences were great among the three populations of cells. Conclusion :The AV/ PI bivariate FCM allows the possibility of early phase of apoptosis in early gast ric carcinoma , which offers a method to research clinically gast ric carcinoma.

Purpose and Methods : In order to screening the antimutagen , the study investigated the mutagenicity and antimutagenicity of 22 kinds of edible chinese herb(Honey , Royal jelly , Bee collecting pollen , Lily , Kely ,Willow bud , the seed of Job’s tear , the Pob of chinese scholort ree , Olired longan , Taro , Sterulaia scaphi , Asiatic plantain , White chrysanthemum , Chinese yam , Coriander , Elm leaf , Pumpkin seeds , Pumpkin pulp , pulp of wagourd , Skin of wagourd Fingered cit ron , Water chestunt by the antimutagenicity and mutagenicity synchronous test . Results and Conclusion : The result s both with and without S9 (rat liver microsomal enzymes) showed that all the samples had no mutagenicity. Honey , Royal jelly , Bee Collecting Pollen , Willow Bud , the pob of chinese scholort ree , Olired longan , White chrysantemum , Chinese yam , Water chestunt were antimutagen against MMC.

Purpose and Methods : The effects of Tripterygiu hypoglaucum(Level) hutch ( THH) on the induction of chromosome abnormalities in human spermatozoa were studied using the human sperm chromosome assay.Results : Result s showed the rates of chromosome aberrations of THH at the doses 50μg/ ml , 100μg/ ml , 200μg/ml were 11. 54 % , 15. 71 % , 16. 79 % respectively , while 7. 25 % in cont rol group. There were significant differences 100μg/ ml group and 200μg/ ml group as compared with cont rol group ( P < 0. 05) . The mean value of chromosome breakages of 100μg/ ml , 200μg/ ml and cont rol groups were 0. 2143 , 0. 2214 and 0. 1346 respectively , the difference was significant ( P < 0. 05) , when the two tested groups compared with the cont rol group.Conclusion : The study result s showed that THH was clastogen in human spermatozoa chromosome aberration test .

Purpose and Methods : To evaluate the P53 protein expression and clinical significance in human hepatocellular carcinoma ( HCC) , the expression of P53 protein was detected in HCC by immunohistochemical method. Results : The result s showed that P53 protein was positive in 49 % (36/ 74) of HCC , and there was significant correlation between P53 protein expression and HCC histologic grade. Conclusion : The results suggested that the over2expression of P53 caused by P53 gene mutation is a common kind of molecular st ructure changes in HCC. In addition , P53 protein may be used as a valuable biomaker of HCC.

Purpose : In order to study the role of anticancer and it s mechanism of acanthopanax sentiocosus (APS) in rat liver during 3′- MeDAB hepato carcinogenesis. Methods : 43 male SD rat s were randomly divided into three groups : group 1. 15 rat s received the basal diet as mormal cont rol. Group 2. 18 rat s received 0. 06 % 3′2MeDAB in there diet for 10 consecutive weeks and then a basal diet . Group 3. 10 rat s received 0. 06 % 3′2MeDAB along with APS (4. 5g/ kg diet) daily in food for 10 weeks. And then a basal diet along with the same dose of APS till the end of experiment . All animals received the diet and water adlibitum. Results :Comparision between two groups was made and the results showed that (1) the content of plasminogen activator inhibitor l( PAI - 1) of serum and liver of APS group rat s was decreased significantly than 3′2MeDAB group ( P < 0. 05) ,(2) The phases of the cell cycle of liver G0/ G phase was increased ( P < 0. 01) , proliferation index ( PI) and DNA index (DI) and the heteroploid were decreased ( P < 0. 01) , Sphase and G2M phase of APS group was decreased significantly than 3′2 MeDAB group ( P < 0. 05) , (3) ras p21 gene protein by FCM was decreased of APS group ( P < 0. 05) , (4) S2P immunohistochemical localization was expression of PCNA and bcl22. The amount of PCNA positive cell were little of APS group. There was no obvious difference of bcl22 expression between two groups , (5) The pathological changes by light and elect ron microscopic observation were light . Conclusion : The present data suggest that APS exhibited a protective effect s in the appearance of cancer induced by 3′ MeDAB through inhibiting the growth of cell decreasing the expression of ras P21 , decreasing the content of PAI2l during hepatocarcinogenesis.

Purpose and Methods : The incidence of micronucleated cells in exfoliated oral mucosa cells and the in2 cidence of micronucleated cells and chromosome aberration in peripheral blood lymphocytes of 54 smoking oral leukoplakia patient s were examined. Results : The result s showed that the incidence of micronucleated cells in exfoliated oral mucosa cells , the incidence of micronucleated cells and chromosome aberration in peripheral blood lymphocytes of smoking oral leukoplakia patient s were significantly higher than those of healthy persons ; the in2 cidence of micronucleated cells in exfoliated oral mucosa cells of lesion mucosa had a positive correlation with the degrees of oral lesions , and the incidence of micronucleated cells in exfoliated oral mucosa cells of normal mucosa had a positive correlation with the incidence of micronucleated cells in peripheral blood lymphocytes. Conclu2 sion : It is concluted that the incidence of micronucleated cells in exfoliated oral mucosa cells of lesion mucosa may reflect the degrees of oral lesions , and it is simple and economical to use the incidence of micronucleated cells in exfoliated oral mucosa cells of normal mucosa to substitute peripheral blood lymphocytes in reflecting DNA damage levels of body exposure to carcinogens.

Purpose and Methods : The effect on antimutation of Lentinan was studied by Ames test and micronucleus test with polychromatic erythrocyte (PCE) of mouse bone marrow in vivo. Results : The rusult s show that the increasing of the number of bacteria in different dose groups can be inhibited by Lentinan. Lentinan inhibited the increasing of micronucleus rate and the inhibition rate is between 45. 32 % and 64. 72 %. Conclusion : It has been observed that Lentinan possess significant antimutagenic effect .

Purpose and Methods :DNA synthesis inhibition (DSI) test was used to study the effect of cooking oil condensate on human amino cells WISH. Results : The result showed that DNA synthesis inhibition rates (DSIR) of cooking oil condensate at 100μg/ ml and 200μg/ ml were 45. 8 % and 85. 8 % respectively , and 0 % at 0. 0μg/ ml in cont rol group. The statistical analysis showed that P value is small than 0. 05. Conclusion : The above2memtioned result s indicated that there were the component s to induce DNA synthesis inhibition in the cooking oil condensate.

Purpose and Methods : The occurrence of apoptosis induced by 8 - Br - cAMP was studied in human Eca - 109 cell line and by means of TUNEL method. Results : The spontaneus apoptosis f requency of human Eca - 109 cells was 10. 3 %. The apoptosis f requency of Eca - 109 cells t reated with 8 - Br - cAMP was 33. 2 %. There was statistical significant difference between the cont rol group and 8 - Br - cAMP induced group. Conclusion : The result s indicated that 8 - Br - cAMP could possess the tendency of increase apoptosis f requency inhuman Eca - 109 cell line.
